| 目的: 观察氧化苦参碱对脂多糖所致小胶质细胞白介素-1β(IL-1β)、肿瘤坏死因子-α
(TNF-α)和白介素-6(IL-6)分泌的影响。方法:予终浓度分别为1、10、20 μg/mL 的氧化苦参碱培养
BV-2 细胞30 min,再加终浓度为1 μg/mL 的脂多糖培养BV-2 细胞30 min、1 h、3 h 和6 h,应用
ELISA 法测定各时间点细胞培养上清液IL-1β、TNF-α 和IL-6 浓度。结果:在30 min、1 h、3 h 和6
h 四个时间点,终浓度为1 μg/mL 的氧化苦参碱不能显著降低细胞培养上清液中IL-1β、TNF-α 和
IL-6 浓度(P>0.05), 终浓度为10、20 μg/mL 的氧化苦参碱剂量依赖性地降低细胞培养上清液中
IL-1β、TNF-α 和IL-6 浓度(P<0.05)。结论:氧化苦参碱可抑制脂多糖所致小胶质细胞炎性因子分
泌。 |

| Objective:To investigate the effect of oxymatrine on the secretion of interleukin-1beta(IL-1β),tumor necrosis
factor-alpha(TNF-α), and interleukin-6(IL-6) in lipopolysaccharide - activated microglia. Methods: Murine BV2 microglial
cells were pretreated with different concentrations of oxymatrine (1,10,and 20 μg/mL) for 30 min followed by
stimulation with lipopolysaccharide(1μg/mL) for different times(30 min, 1 h, 3 h, and 6 h). Concentrations of TNF-α,
IL-1β, and IL-6 in supernatant were determined using ELISA. Results:Oxymatrine of 1μg/ml did not significantly attenuate
the productions of TNF-α, IL-1β and IL-6(P>0.05),while 10μg/mL and 20μg/mL oxymatrine suppressed their productions
in lipopolysaccharide - stimulated BV2 microglial cells in a dose-dependent manner(P<0.05). Conclusions:
Oxymatrine may suppress the secretion of inflammatory cytokines in lipopolysaccharide - activated microglial cells. |
