      目的: 探讨万寿菊提取物对大鼠视网膜光损伤过程中细胞凋亡因子半胱氨酸蛋白酶-3 (Caspase-3) 的影响。方法: SD大鼠40只, 随机分为正常对照组、 模型对照组、 万寿菊提取物低剂量 组与高剂量组, 各10只。除正常对照组外, 其余3组大鼠均缝成开睑, 采用日光灯照射, 照光强度 (3000±100) Lux, 持续12h。光照射完毕后立即拆除缝线送回暗环境饲养笼中饲养12h, 再重复以上 过程, 连续3次, 光照射时间总计36h, 建立视网膜光损伤模型。各组在光照前充分暗适应36h, 暗 适应前万寿菊提取物低剂量组、 高剂量组大鼠给药, 1天1次, 直至光照后7天。实验结束, 处死大 鼠, 检测视网膜细胞中Caspase-3表达情况。结果: 光照后模型对照组大鼠视网膜外核层细胞浆中 Caspase-3阳性物质弥漫分布, 含量增多。其面积、 平均光密度、 积分光密度和平均黑度均高于正 常对照组 (P<0.05), 而万寿菊低剂量治疗组视网膜外核层细胞浆中Caspase-3阳性物质减少明显, 四项指标与模型对照组比较, 差异有统计学意义 (P<0.05), 高剂量治疗组阳性物质减少不明显。 结论: 万寿菊提取物对大鼠视网膜光损伤有修复作用, 其作用机制可能与减少Caspase-3的表达, 从而拮抗细胞凋亡有关。

      Objective: To explore the influence of marigold extract on the expression of caspase-3 in retinal light damage. Methods: Forty SD rats were randomly divided into 4 group: normal control group, model group, lose-dose marigold extract group, and high-dose marigold extract group, 10 in each. To establish the retinal pho? tic injury model, except the normal group, other 3 groups received light irritation with light intensity of 3000± 100 Lux by fluorescent lamps for 12h and repeated 3 times in a row. Before the light explosion, all rats had dark adaption for 36 h;marigold extract was given of low dose and high dose once a day at the same time and the medication lasted till 7 days after light irritation. Then the rats were sacrificed to detect caspase-3 in reti? nal cells. Results: Compared with the normal control group, the content of caspase-3 positive substance in the retinal outer nuclear layer plasma of the model group was greater, with larger area, higher mean optical density, integral optical density and mean degree of darkness(P<0.05). Compared with the model group, the content of cas? pase-3 positive substance and other 4 parameters in the low-dose and high-dose marigold extract groups were statically less(P<0.05), but no significant difference was noted between the 2 marigold extract groups(P>0.05). Conclusion: Marigold extract can repair the retinal photic injury by reducing the expression of caspase-3 to an? tagonize cell apoptosis.
