目的：观察苦参碱对人乳腺癌Bcap-37细胞增殖的影响，并探讨其作用机制。方法：用 MTT 法检测苦参碱对Bcap-37细胞的抑制率，流式细胞仪检测细胞凋亡率，Western blot法检测 Caspase-3、Bax、Bcl-2蛋白表达。结果：苦参碱对Bcap-37细胞增殖具有明显抑制作用，且具有剂量和时间依赖性。与对照组比较，药物作用24、48、72h后，苦参碱组的凋亡率均明显增高（P< 0.01），并以作用 48h 最明显。Caspase-3、Bax 蛋白表达增高，Bcl-2 蛋白降低。结论：苦参碱对 Bcap-37细胞生长具有明显抑制作用，其作用机制可能与上调Caspase-3、Bax蛋白，下调Bcl-2蛋白表达，促进细胞凋亡有关。

英文摘要:

Objective: To observe the effect of matrine on the growth of human breast cancer cell line Bcap-37 and the underlying mechanism. Methods: Bcap-37 cell line were treated with matrine at doses of 78.125, 156.25, 312.5, 625.0, 1250.0, 2500.0, and 5000.0mg/L. The cell inhibitory rate was evaluated by MMT method. The cell apoptosis rate was detected with flow cytometry. The expression of caspas-3, Bax, and Bcl-2 was mea? sured by Western blotting. Results: Matrine markedly suppressed the growth of Bcap-37 cells in a dose-depen? dant manner. Compared with the control group, the cell apoptosis rate in 24, 48, and 72h after matrine treat? ment was increased(P<0.01) and the apoptosis reached the peak in 48h. Western blotting analysis showed that the expression of caspase-3 and Bax were elevated while Bcl-2 was reduced. Conclusion: Matrine may have an inhibitory effect on the growth of Bcap-37 cells. The inhibitory effect may be attributed to the increase in cas? pase-3 and Bax and the decease in Bcl-2.
