      目的:观察三七总皂苷对肝素结合性表皮生长因子样生长因子(HB-EGF)诱导大鼠肾小 球系膜细胞增殖的作用。方法:以原代培养大鼠肾小球系膜细胞为细胞模型,以400ng/mL 三七总 皂苷和100ng/mL HB-EGF为干预因素,实验分对照组、100ng/mL HB-EGF组和400ng/mL三七总 皂苷+100ng/mL HB-EGF组,刺激系膜细胞24、36、48h,采用MTT方法,观察三七总皂苷对重组 HB-EGF诱导系膜细胞增殖的作用情况。结果:HB-EGF刺激36h 时,系膜细胞明显增殖,与对照组 比较,P<0.05;三七总皂苷能抑制HB-EGF诱导的系膜细胞的增殖,与HB-EGF组比较,P<0.05。随 着刺激时间的延长,三七总皂苷对HB-EGF 诱导系膜细胞增殖的抑制作用增强,但36h 与24h 比 较,差异无统计学意义(P>0.05);与48h 比较,差异有统计学意义(P<0.05)。结论:三七总皂苷能抑 制HB-EGF诱导的系膜细胞增殖作用,且作用随着刺激时间延长而增强。三七总皂苷可能通过抑 制HB-EGF对系膜细胞增殖作用而延缓肾小球硬化。
英文摘要:
      Objective:To investigate the influence of panax notoginseng saponins(PNS)on proliferation of mesangial cell induced by heparin-binding epidermal growth factor like growth factor(HB-EGF)and its mechanism in kidney disease. Methods:The primary cultured SD rat mesangial cell were used as cell models. The cells were divided into control group,100 ng/ml HB-EGF group,and 400 ng/ml PNS+100 ng/ml HB-EGF group,and were cultured with these solutions for 24,36,and 48 h,respectively. Cell proliferation was assessed by MTT colorimetric assay. Results:HB-EGF stimulated the proliferation of mesangial cells in 36 h(compared with control group P<0.05)but the effect was inhibited by PNS(compared with HB-EGG group P<0.05),and the effect of inhibition in 48 h was more obvious than that in 36 h(P<0.05). But no significant difference in the effect of inhibition was noted between cells treated for 36 h and 24 h(P>0.05). Conclusion:PNS can inhibit the proliferation of mesangial cells induced by HB-EGF,and the effect of inhibition gradually increases with the prolongation of the culture time. PNS may re- tard the development of glomerular sclerosis by inhibiting the proliferation of mesangial cells induced by HB-EGF.
