      目的:研究柿叶黄酮对同型半胱氨酸诱导的小胶质细胞表达白介素-1β(IL-1β)、白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)的影响,并将其结果与银杏叶提取物EGb761进行比较。方法:将体外培养的小鼠小胶质瘤细胞(BV-2细胞)分成空白对照组、同型半胱氨酸组、EGb761组(100mg/L)和柿叶黄酮低、中、高(12.5、25、50ug/mL)剂量组,培养72h。应用RT-qPCR方法评价IL-1β、IL-6和TNF-α mRNA表达,酶联免疫吸附法(ELISA)测定培养上清中上述细胞因子的蛋白浓度。结果:与对照组相比,同型半胱氨酸组细胞内IL-1β、IL-6和TNF-α mRNA表达和细胞培养上清液中蛋白浓度明显增加;与同型半胱氨酸组相比,EGb761组和中、高剂量柿叶黄酮组各细胞因子mRNA表达及培养上清液中蛋白浓度均降低,尤其是高剂量柿叶黄酮组结果与EGb761组作用相似。结论:柿叶黄酮能抑制同型半胱氨酸诱导小胶质细胞表达IL-1β、IL-6和TNF-α,在一定浓度条件下其作用不亚于银杏叶提取物。

      Objective: To evaluate the effect of flavone from diospyros kaki on the expression of interleukin-1β(IL-1β), interleukin-6(IL-6) and tumor necrosis factor-α(TNF-α) in culturing for microgia (BV-2 cells) induced by homocysteine. Then the effect was compared with that of EGb761. Method: The BV-2 cells incubated vitro were divided into six groups: blank control group, homocysteine(Hcy) group, EGb761 group(100mg/L) and flavone from diospyros kaki groups of different concentration(12.5, 25, 50ug/mL).The mRNA expression of IL-1β, IL-6 and TNF-α was assessed by RT-qPCR.The supernatant protein was detected by ELISA. Result: The mRNA expression and supernatant protein of IL-1β, IL-6, TNF-α in Hcy group were significantly higher than those of control group, but were decreased significantly in EGb761 group and flavone from diospyros kaki groups. Especially, both the mRNA expression and supernatant protein in higher concentration flavone from diospyros kaki group were comparable to those in EGb761 group.Conclusion:Flavone from diospyros kaki can inhibit the expression of IL-1β, IL-6 and TNF-α in cultured BV-2 cells induced by homocysteine. In a certain concentration, it was no less effective than EGb761.
