| 目的:探讨百草枯中毒致肺纤维化大鼠肺组织中TGF-β1、HIF-1α的表达以及茶多酚(Tea Polyphenols,TP)及黄芩(scutellariabai calensisgeorgi,SC)对急性百草枯中毒肺纤维化的干预作用。方法:90只健康SD大鼠随机分为对照组(N组)、PQ组(P组)、PQ TP组(T组)、PQ SC组(S组)及PQ TP SC组(L组),每组18只,P、T、S、L各组大鼠予20%PQ 50mg/kg一次性灌胃染毒;大鼠染毒6小时后T组给予茶多酚250 mg/kg.d、S组给予黄芩80 mg/kg.d、L组给予茶多酚及黄芩复合制剂灌胃干预,N组和P组同时用等量生理盐水灌胃。在第1d、7d和14d随机处死大鼠,腹主动脉采血,采用酶联免疫吸附法(ELISA)检测血清TGF-βl、HIF-1α水平,免疫组化染色检测大鼠肺组织切片TGF-βl、HIF-1α水平。数据采用( ±S)表示,应用SPSS 20.0软件进行统计学分析。结果:PQ 染毒后P组血清及肺组织中TGF-βl、HIF-1α含量高于N组 (p <0. 05),经中药干预后,各治疗组血清中TGF-βl、HIF-1α水平下降,差异有显著性(p <0. 05)。相比于对照组,百草枯中毒组(P组)、中药干预组(T、S及L组)肺组织中TGF-βl、、HIF-1α表达于第7 d 、14d升高,差异有统计学意义(p < 0.05),P组表达量最高,中药干预组(T、S及L组)第7 d和14 d时TGF-βl、HIF-1α表达较P组降低(p < 0.05)。各中药干预组各时间TGF-βl、HIF-1α表达差异无统计学意义(p > 0.05)。结论:茶多酚、黄芩提取物可使急性PQ 中毒引起的肺纤维化得到改善。 |

| Objective:To study the Changes of TGF-β1 and HIF-1αcaused by acute paraquat poisoning in rat and protective effect of Tea Polyphenols(TP)and scutellariabaicalensisgeorgi(SC).Method:Ninty healthy Sprague-Drawley(SD) rats were randomly divided into Control group(group N)、PQ group(group P)、 PQ TP group(group T)、PQ SC group(group S)and PQ TP SC group(group L. Group P、T、S、L were treated intragastrically with 20% PQ at 50mg/kg at a time, Group T was given TP at 250mg/kg.d,Group S was given SC at 80mg/kg.d and Group L was given TP and SC at 6h after oral administration of PQ, while only saline was treated intragastrically the other two groups. Rats were sacrificed to obtain blood from abdominal aorta on the 1st, 7th, and 14th day. Then the levels in serum of TGF-βl and HIF-1αwere detected by ELISA kits.The lungs were taken for the expression of TGF-βl and HIF-1α protein by immunohistochemistry. Data were expressed as mean±standard error of the mean( ±S).Statistical analysis was carried out with the soft SPSS 20.0. Results: Compared with group N, the levels of TGF-βl and HIF-1αin the group P increased significantly (p < 0.05) both in serum and lung tissue; After drug intervention ,the serum levels of TGF-βl、HIF-1α in the group P、T、S、L were lower than that of group P (p < 0.05); Compared with group N,The expression levels of TGF-βl、HIF-1αin lung tissues at 7th d and 14th d significantly increased(p < 0.05), Group P was the highest level. Compared with Group P, The expression levels of TGF-βl、HIF-1αin lung tissues in the group T、S、L decreased(p < 0.05). but there was no statistical difference between the group T、S、L both on the 1 st d、7th d and 13th d (p > 0.05).Conclusions:Administation of TP and SC could improve the pulmonary fibrosis of rats caused by acute paraquat poisoning. |
