| 目的:研究绞股蓝总皂苷对糖尿病肾病大鼠(DN)肾脏nephrin、VEGF表达的影响,进一步探讨其降低尿蛋白、保护肾脏的机理。方法:采用单侧肾切除加链脲佐菌素(STZ)注射法改良复制DN模型,实验分为正常组、模型组、治疗组(绞股蓝总皂苷高、中、低剂量组和缬沙坦组)。药物干预4周后测定24小时尿蛋白、血肌酐、尿素氮及肾脏肥大指数,免疫组化测定肾小球nephrin、VEGF蛋白的表达水平。结果:1.绞股蓝高剂量组24h尿蛋白、血肌酐、尿素氮、肾脏肥大指数明显低于模型组(P均<0.05),但低剂量组无差异,(P>0.05);2. nephrin 蛋白表达水平较模型组明显上调(P<0.01),VEGF表达明显抑制(P<0.05),其中高剂量组和缬沙坦组效果类似,明显优于低剂量组(P<0.01)。结论:绞股蓝总皂苷可上调足细胞相关分子nephrin蛋白的表达,抑制足细胞分泌的VEGF过表达,降低24小时尿蛋白,改善肾功能异常,从而保护足细胞,延缓肾小球硬化。 |

| Objective: To investigate the effect of gypenoside on the expressions of nephrin, vascular endothelial growth factor (VEGF) in rats with diabetic nephropathy (DN) and explore its mechanism of the reduction of proteinuria and renal protection. Methods: DN model was established by uninephrectomized and induced with streptozotocin (STZ), then randomly divided into model group and treatment groups (groups of high, medium, low dose of gypenoside, and valsartan group), with normal rats as the control group. After the treatment for 4 weeks, check out the renal function, 24h urine volume and 24h urinary protein, measure the glomerular hypertrophy. The expression of nephrin、VEGF protein was examined by Immunohistochemistry.Result:1. The levels of 24h urinary protein count、BUN、SCr and Kidney hypertrophy index all significantly decreased in group of high gypenoside( P<0.05),but no difference in group of low gypenoside(P>0.05);2. the expression of nephrin in treatment groups were upregulated as compared to control group ( P <0.01), while VEGF expression signi fi cantly inhibited ( P <0.01). The result in high dose group was much better than low dose group ( P<0.01) and similar to valsartan group.Conclusion: Gypenoside can provide renal protection, reduce proteinuria against glomerulosclerosis in rats by reducing VEGF in expression and increasing nephrin expression the podocytes. |
