| 目的:研究三生合欢饮抗抑郁作用及其对肠道菌群的影响 方法:健康雄性昆明种小鼠60只,随机分为6组,即空白组、模型组、百忧解组及三生和欢饮高、中、低剂量组。采用慢性应激和孤养的方法复制慢性应激抑郁大鼠模型。第21天测定小鼠体重增加情况、糖水消耗、行为学以及脑内多巴胺递质含量等指标。第22天,采集各实验组小鼠粪便样品,采用ERIC-PCR技术获得反映肠道微生物群落结构特征的ERIC-PCR指纹图谱。结果:与空白组比较,模型组鼠在第14天后体重增加已有显著降低趋势,第21天后的体重增加也具有显著意义(P<0.01), 与模型组相比,三生合欢饮高剂量组、百忧解组小鼠体重增加明显高于模型组(P<0.01); 与空白组比较,模型组小鼠在第14天后的糖水消耗有显著降低趋势(P<0.01),第21天后也有明显降低(P<0.05)。与模型组比较,三生合欢饮高﹑中﹑低剂量组小鼠糖水消耗量显著增加(P<0.01),百忧解组小鼠糖水消耗亦明显增加(P<0.05); 与空白组比较,模型组小鼠水平活动和垂直活动次数明显减少(P<0.05),游泳不动时间明显增加(P<0.05);与模型组比较,三生合欢饮低、中剂量组小鼠自主活动次数显著增加(P<0.01),高剂量组小鼠游泳不动时间明显减少(P<0.05);与空白组比较,三生合欢饮模型组DA递质的含量显著少于空白组(P<0.05);与模型组比较,三生合欢饮低剂量组对DA含量也有明显提高(P<0.05)。
结论:三生合欢饮有较好的抗抑郁作用,并能调节抑郁模型小组肠道菌群,使之趋于正常。 |

| Objective: To investigate the Antidepressant effect and its effect on intestinal flora of Sanshenghehuan decoction. Methods: 60 healthy male mice, divided into 6 groups randomly: blank control group, model group, Fluoxetine group, Sanshenghehuan decoction groups (high, medium and low dose).Chronic depression rat model was induced by chronic stress and isolated breeding. Weight gain, the autonomous activity, and monoamine neurotransmitter were determination. Collect fecal samples in each experimental group of mice, obtained by ERIC-PCR technique to reflect intestinal microbial community structure of ERIC-PCR fingerprinting. Results:Compared with blank group,model group rats gain weight has been significangtly reduced trend after 14 days,21 days also has the remarkable significance(P<0.01),Compared with model group, high dose group ,fluoxertine hydrochloride group gain weight in mice significantly higher than the model group(P<0.01); . Compared with blank group ,model group mice has significantly lower in 14 days after the sugar water consumption trend(P<0.01),after 21 days also have significantly lower(P<0.05).Compared with model group ,high dose ,middle dose, low dose group mice increased significantly(P<0.01), ,sugar water consumption of fluoxertine hydrochloride group also increased significantly(P<0.05).Compared with the blank group,the number of horizontal and vertical activities of model group mice decreased significantly(P<0.05);Compared with model group ,the number of autonomic activities in mice of low、medium dose group increased significantly(P<0.01);Compared with the blank group, DA transmitters of model group is significantly less than blank group(P<0.05);Compared with model group, the DA transmitters of low dose has increased significantly.
Conclusion: Sanshenghehuan decoction has some antidepressant effect, by improving the intestinal flora to the treatment of depression. |
