      【】目的:观察姜黄素(curcumin, cur)对人支气管上皮细胞(human bronchial epithelial cells, HBECs)MMP-9表达的影响。方法:体外培养人支气管上皮细胞,分为空白组、LPS组及姜黄素组,其中姜黄素组在相同浓度LPS刺激下又各分为5、10、20、40和80μmol/L 5个浓度组,作用4h后在显微镜下观察cur干预后细胞的形态变化,采用实时荧光定量PCR观察人支气管上皮细胞MMP-9 mRNA的表达变化。结果:显微镜下观察姜黄素干预后的细胞形态良好,细胞无明显损伤。实时PCR结果显示,空白组可见一定量MMP-9 mRNA表达,与空白组比较,LPS组的人支气管上皮细胞MMP-9 mRNA表达量明显增加,80μmol/L cur亦可增加MMP-9 mRNA的表达(p<0.05)。与LPS组比较,10μmol/L cur组人支气管上皮细胞的MMP-9 mRNA表达量显著下降(p<0.05),5μmol/L、20μmol/L、40μmol/L的cur对支气管上皮细胞MMP-9的mRNA表达无明显作用。结论:10μmol/L姜黄素可降低人支气管上皮细胞的MMP-9mRNA的表达。

      Objective:To observe the role of curcumin (cur) on the expression of MMP-9 in human bronchial epithelial cells(HBECs). Methods:HBECs was cultured in vitro and divided into blank group,LPS group and cur groups,and cur groups was set at 5 concentrations of 5,10,20,40,80μmol / L after the stimulation of the same LPS concentration .Observe the morphological changes of cells through a microscope after cur intervention and real-time quantitative PCR was performed to examine the dynamic expression of MMP-9 in human bronchial epithelial cells after 4 hours.Results:The cells’ morphology were well without significant damage after cur intervention by means of the microscope.The consequences of real-time PCR show that the blank group could produce a certain amount of MMP-9 expression,and compared with it,the group of LPS has a significantly increasing in the expression of MMP-9 in the human bronchial epithelial cells as well as the subgroup of 80μmol/L cur (p <0.05).Compared with the group of LPS,the concentration subgroup of 10μmol / L cur’ expression of MMP-9 in human bronchial epithelial cells decreased significantly(p <0.05), the concentration of 5μmol / L, 20μmol / L, 40μmol / L of cur has no marked effect.Conclusion:10μmol/L of Curcumin can lower the expression of MMP-9 in the human bronchial epithelial cells.
