      目的探索消乳块胶囊对大鼠乳腺肿瘤血浆胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)的影响。方法50天龄Sprague-Dawley系健康雌性未孕大鼠64只随机分为3组:模型组、治疗组(各25只)和对照组(14只)。模型组和治疗组大鼠均先经两次9,10-二甲基苯蒽(9,10-dimethylben(a) Anthracene ,DMBA 15mg溶于1.5ml芝麻油, 间隔1周)灌胃造模,同时治疗组大鼠以消乳块胶囊 0.4g/kg/d灌胃,持续给药,在第11周和14周分批处死。大鼠乳腺组织及肿瘤组织均作常规病理,酶联免疫吸附法测定IGF-1水平。结果 同正常组相比,癌变及增生组血浆IGF-1水平升高明显(P<0.01),尤其在癌变组升高更为明显,同增生组相比,差异仍有显著性(P<0.01)。血浆IGF-1测定显示,模型组、治疗组较对照组有明显增高,差异有统计学意义(P<0.01), 治疗组较模型组有明显下降,差异同样有统计学意义(P<0.01)。结论 消乳块胶囊对DMBA诱导的大鼠乳腺恶性肿瘤有部分预防作用,其机制可能是通过降低血浆IGF-1水平来实现。
英文摘要:
      ABSTRACT Objective To investigate the anticancer action of “Xiao RuKuai” capsuleon in rat’s mammary tumor.Methods 50 days old female Sprague-Dawley rats were randomly divided into 3 groups: model group, therapy group and negative control group. The model group and therapy group were given DMBA by intragastric administration ( 15 mg in 1.5 ml sesame oil, twice, one week interval). At the same time, the rats in the therapy group were given the “XiaoRuKuai” capsule (0.4g/kg/d)by intragastric administration. The rats were killed in 11th or14th week. The breast tissue or neoplasma were observed by routine pathology ( HE stain). The plasma IGF-1 levels were detected by ELISA assay. Results Compared to the normal group ,The levels of IGF-1 were remarkably higher in the Cancerous and hyperplastic group( P< 0.01), Especially in the canceration group, there is still a significant difference compared with hyperplasia group (P < 0.01) . For the levels of IGF-1 in the plasma , we found that they were remarkably higher in the model and therapy group comparing the control group ( P<0.01). After we gave the “Xiao RuKuai” capsule, IGF-1 levels had going down remarkably, Compared to the model group, there is still a significant difference (P < 0.01) . Conclusion The “Xiao RuKuai” capsule has partly efficacy on preventing the mammary carcinogenesis induced by DMBA .The mechanism is probably associated with regulating IGF-1 levels.
