| 目的:观察丹红注射液对脓毒症大鼠心肌损伤的保护作用并探讨其可能机制。方法:雄性Wistar大鼠分为4组:正常组、假手术组、模型组、丹红组,每组各8只。盲肠结扎并穿孔(CLP)法建立脓毒症大鼠动物模型,观察各组心肌组织超微结构改变,心肌组织丙二醛(MDA)、超氧化物岐化酶(SOD)和高迁移率族蛋白1(HMGB1)含量的变化。结果:CLP术后24h,模型组大鼠心肌组织MDA含量、HMGB1蛋白合成升高、SOD活力则降低,与假手术组比较有差异(P<0.05),丹红组较模型组均有改善,改善有统计学差异(P<0.05);超微结构示模型组大鼠心肌细胞线粒体肿胀,部分空泡变性;丹红组心肌细胞损伤轻于模型组同时间点。结论:丹红注射液对脓毒症大鼠心肌损伤有一定程度的保护作用,其机制可能与清除氧自由基,降低HMGB1含量有关。 |

| Objective: To investigate whether Danhong Injection has a protective effect on myocardial injury in rats with cecal ligation and puncture(CLP)-induced sepsis,and observe the mechanism of the protective effect. Methods: Wistar rats were randomly devided into four groups: Normal group、Sham-operated group、Model group、and Danhong Injection group. There were 8 rats in every group. To make sepsis rat model by cecal ligation and puncture. The difference of ultrastructure of myocardium cell were observed and contrasted in every group. At the same time, the following item in heart tissue was observed: the Malondialdehyde (MDA) contents、the Superoxidedismutase(SOD) contents、and the protein synthesis of High mobility group box-1 protein(HMGB1). Result: 24 hours after CLP, The MDA contents and the protein synthesis of HMGB1 in Model group’s heart tissue were increased, while the SOD contents decreased, the changes were different from them in Sham-operated group (P<0.05). Compared with Model group, Danhong Injection group had lower MDA contents , lower HMGB1 contents and higher SOD contents (P<0.05). The mitochondrion of heart cell in Model group got swelling, most of heart cell autopepsia. Above changes were not more striking in Danhong groups than in Model groups. Conclusion: Our results suggest that Danhong Injection can protect against myocardial injury in sepsis, this effect is related to scavenging oxygen-free radicals and reducing the protein synthesis of HMGB1. |
