| 目的 探讨冬虫夏草提取物对混合淋巴细胞(MLC)培养体系Treg/Th17轴的影响,明确冬虫夏草提取物的免疫调节机制。方法 体外建立单向混合淋巴细胞培养体系,用冬虫夏草提取物干预培养体系,在不同的时间点收集细胞,采用流式细胞术检测细胞中辅助性T细胞17(Th17细胞)和CD4+CD25+T调节性T淋巴细胞(Treg细胞)的比例,并计算出Th17细胞与Treg细胞的比值;采用酶联免疫吸附试验法分别检测培养液中白细胞介素(IL)-17、IL-23、IL-2和IL-6的浓度。结果 冬虫夏草提取物干预组12、24、36h三个时间点T细胞中Treg细胞比例分别为13.17±1.25、12.3±1.31、11.89 ± 1.46(%),同时间点的对照组T细胞中Treg细胞比例分别为11.04±1.32、10.16±1.15、8.91±0.97(%),差异均有统计学意义(P<0.05);干预组T细胞中Th17细胞比例分别为2.81 ± 0.13、4.02±0.38、4.95±0.49(%),同时间点的对照组T细胞中Th17细胞比例分别为4.65± 0.31、6.63±0.63、8.03±0.76(%),差异均有统计学意义(P<0.05)。冬虫夏草提取物干预组12、24、36h三个时间点T细胞中Th17细胞与Treg细胞的比值均显著低于同时间点的对照组(P<0.05)。冬虫夏草提取物干预组12、24、36h三个时间点培养液中IL-2水平明显高于同时间点的对照组(P<0.05),而干预组培养液中IL-6、IL-17、IL-23水平低于同时间点的对照组(P<0.05)。 结论 冬虫夏草提取物能够明显促进MLC体系细胞中Treg细胞增殖,抑制Thl7细胞增殖,降低Th17/Treg细胞比值,改变Th17/Treg轴的平衡,同时增加IL-2的分泌、减少IL-6、IL-17、IL-23的分泌,有利于诱导免疫耐受。 |

| Objective To investigate the influence of Cordyceps sinensis extract on the Treg/Th17 axis of mixed lymphocyte culture (MLC) system ,and investigate the immune regulation mechanism of Cordyceps sinensis extract. Method Establish one-way mixed lymphocyte culture system in vitro, Use of Cordyceps sinensis Extract intervention system, cells were collected at different time points, flow cytometry was used to detect Th17 cells and Treg cells, and calculate the ratio of TH17 cells and Treg cells; Enzyme linked immunosorbent assay was used to
detect concentrations of medium interleukin (IL) -17, IL-23, IL-2 and IL-6. Result The ratio of Treg cell in Cordyceps sinensis extract treatment group were 13.17±1.25、12.3±1.31、11.89 ± 1.46(%)at 12、24、36h, the ratio of Treg cell in the control group were 11.04±1.32、10.16±1.15、8.91±0.97(%)at the same time point, the difference was statistically significant (P<0.05); the ratio of Th17 cells in Cordyceps sinensis extract treatment group were 2.81 ± 0.13、4.02±0.38、4.95±0.49(%)at 12、24、36h, the ratio of Th17 cell in the control group were 4.65± 0.31、6.63±0.63、8.03±0.76(%)at the same time point, the difference was statistically significant (P<0.05). The ratio of TH17 /Treg in Cordyceps sinensis extract group were significantly lower than that of the control group at the same time points (P<0.05). IL-2 levels of medium in Cordyceps sinensis extract group were significantly higher than those in the control group at the same time points (P<0.05), while IL-6, IL-17, IL-23 levels were significantly lower than those in the control group at the same time points (P<0.05). Conclusion Cordyceps sinensis extract can obviously promote the proliferation of Treg cells, inhibition the proliferation of Thl7 cell, reduce the ratio of Th17/Treg, and change the balance of Th17/Treg axis in the system of MLC cells. And it can increasing the secretion of IL-2, reduce the secretion of IL-6, IL-17, IL-23, which is advantageous to the induction of immune tolerance. |
