      目的 研究槐耳清膏对人胃癌MKN-45细胞在裸鼠体内生长和转移的抑制作用,并探讨其作用机制。方法 采用BALB/c裸鼠建立人胃癌MKN-45细胞淋巴结转移模型,随机分为槐耳清膏组、空白对照组,每组各20只。建模后,每5天测量裸鼠荷瘤并计算肿瘤相对体积(RTV);用药5周后处死裸鼠,Real-time qPCR检测肿瘤组织中Wnt、β-catenin、E-cadherin、N-cadherin和VEGF表达水平。结果 (1)槐耳清膏组末次测得的足垫瘤RTV为(662.75±450.66),明显小于空白对照组的RTV(1611.85±611.83)(P<0.01)。(2)槐耳清膏组淋巴结转移率15/20(75%)与对照组8/20(40%)相比,其差异有统计学意义(P<0.05)。(3)槐耳清膏组肿瘤组织中Wnt、β-catenin、N-cadherin和VEGF表达低于空白对照组,而E-cadherin的表达高于对照组(P<0.05)。结论 槐耳清膏可抑制人胃癌MKN-45细胞在裸鼠上的生长及转移,其机制可能与抗血管生成和阻碍上皮间质转化(EMT)过程有关。
英文摘要:
      Objective To study the effects of trametes robiniophila on the human MKN - 45 gastric cancer cells inhibition of growth and metastasis in nude mice, and explore the mechanism.Methods BALB/c nude mice were inoculated with MKN-45 cells to establish the lymph node metastasis model of human gastric carcinoma. After the model was successfully established, 40 mice were randomly divided into 2 groups (n=20 in each group): control group, trametes robiniophila group. After the mass formatted,the caliper was used to measure the tumor size every five days. The Relative tumor volume (RTV) was calculated. After 5 weeks, the nude mice were killed Wnt, beta - catenin, N – cadherin, VEGF mRNA expression were detected by Real-time qPCR. Results (1)After 5 weeks of drugs intervention, tumor relative volume (RTV) in trametes robiniophila group were (662.75±450.66),which was less than that in control group (1611.85±611.83) (P<0.01). (2) The lymph node metastasis rate of trametes robiniophila group 8/20 (40%) compared with that in control group 15/20 (75%) was significantly decreased (p<0.05). (3)Compared with that in control group, Wnt, β-catenin, N-cadherin, VEGF mRNA in trametes robiniophila group were down-regulated while E-cadherin mRNA expression were up-regulated (P<0.05). Conclusion Trametes robiniophila could inhibit the growth and lymphatic metastasis of human gastric cancer in nude mice, which might be related with anti-angiogenesis and setting the Epithelial-mesenchymal Transition (EMT) process back.
