      目的 明确水飞蓟素(Silymarin,Sily) 对人乳腺癌耐药细胞株MCF-7/ADM的逆转耐药作用。方法 以CCK-8法测定阿霉素(Adriamycin,Adm)对人乳腺癌敏感细胞株MCF-7/S和耐药细胞株MCF-7/ADM的毒性作用,计算出耐药倍数。以无细胞毒性的Sily(10μg/ml)作为逆转耐药剂,联合Adm观察其对耐药细胞株MCF-7/ADM的逆转耐药作用,计算得逆转倍数。结果 ①Adm对MCF-7/S和MCF-7/ADM的半数抑制浓度(IC50)分别为1.773 μg/ml和43.812 μg/ml,耐药倍数为24.7倍。②Sily能够增强ADM对MCF-7/ADM的细胞毒作用。以10μg/ml(抑制率为2.0%)的Sily联合Adm作用于MCF-7/ADM 48h后,耐药细胞株的IC50降至7.798 μg/ml,逆转倍数为5.6倍(P <0.01)。结论 Sily能够逆转人乳腺癌耐药细胞株MCF-7/ADM的耐药性。
英文摘要:
      Objective To explore the effect of Silymarin on Adriamycin resistance of breast cancer cell line. Methods To measure toxicant effect of Adm to MCF- 7/S (sensitive strain) and MCF-7/ADM (multidrug- resistant strain) for resistant index by CCK-8 in vitro ; to observe the reversal effect of Silymarin (10μg/ml) which has screened for non-toxicant to MCF- 7/ADM by CCK-8. Results ① IC50 of Adm to MCF-7/S and MCF-7/ADM were 1.773 μg/ml and 43.812 μg/ml respectively. Drug-Resistant index of MCF-7/ADM was 24.7. ② Silymarin could increase cell toxic effect of Adm. After treated with 10μg/ml silymarin (Inhibition rate was 2.0%) combining with Adm to MCF- 7/ADM for 48 hours , cell’s IC50 declined to 7.798 μg/ml, and reversal index is 5.6 (P < 0.01 ). Conclusion Silymarin could reduce partly reverse multidrug- resistance to Adm of MCF- 7/ADM in vitro.
