| 目的: 探讨复方大黄膏的抗炎镇痛作用及其可能机制。方法: 采用二甲苯致小鼠耳廓肿胀法观察该药的消肿作用；采用热板法及醋酸扭体法观察该药的镇痛作用; 采用Western blotting检测耳廓组织TNF-α、IL-1β蛋白的表达以研究药物的可能抗炎镇痛机制。结果：与赋形剂对照组比较,复方大黄膏低、中、高剂量组的小鼠耳廓肿胀明显减轻,中、高剂量组在用药后30min、60min、120min小鼠的痛阈值明显提高,小鼠扭体次数明显减少,低剂量组在用药后30min、60min的痛阈值明显提高,低、中、高剂量组耳廓组织的TNF-α和IL-1β蛋白表达均明显降低。结论: 复方大黄膏具有良好的抗炎镇痛作用,其机制可能与抑制组织TNF-α和IL-1β等炎症介质的表达有关。 |

| Objective: To investigate anti-inflammatory and analgesic effects of Fufang dahuang ointment and its mechanism.Method:The anti-inflammatory effect was evaluated by dimethylbenzene-caused ear swelling.The hotplate test and the acetic acid writhing in mice were used to study the analgesic action．The expression of TNF-αand IL-1βin the inflammatory auricula tissue was assessed by Western blotting.Ｒesult: Compared with control group, the mice ear swelling in the low, middle and high doses group of Fufang dahuang ointment were significantly inhibited. The middle and high doses of Fufang dahuang ointment not only obviously improve the pain threshold of hot-plate test at 30min,60min and 120min after drug administration,but also obviously decrease writhing times injected with acetic acid in mice．The low dose of Fufang dahuang ointment remarkably improve the pain threshold of hot-plate test at 30min and 60min after drug administration. Furthermore, Fufang dahuang ointment in low, middle and high doses group obviously down-regulated the protein expression of TNF-αand IL-1βin the inflammatory tissue．Conclusion: Fufang dahuang ointment has remarkable anti-inflammatory and analgesic effects,which may be related to the inhibition of inflammatory mediators such as TNF-αand IL-1β． |
