| [摘要]:目的 探讨中药蟾酥作用的不同时间段对多重耐药鲍曼不动杆菌氨基糖苷类耐药相关基因表达的影响。方法 利用RT-PCR方法检测两种蟾酥提取液作用不同时间段对多重耐药鲍曼不动杆菌氨基糖苷类抗菌药物相关耐药基因表达的变化。结果 鲍曼不动杆菌菌株与醇提蟾酥和水提蟾酥分别作用16到24小时后,氨基糖苷类耐药相关耐药基因aac( 6') -II和aac( 6') -Ⅰ的表达水平均有不同程度下降,与其他时间段的结果相比,差异有统计学意义(<0.05);两种蟾酥制剂对AMI-aphA1和AMIaadA1基因表达的抑制作用基本一致,且作用2小时后基本无抑制作用。醇提蟾酥作用16小时,对ant(3〞)-Ⅰ耐药基因表达有微弱抑制作用。两种制剂作用相同时间后比较CT值,aac( 6') –II、aac( 6') -Ⅰ、 ant(3〞)-Ⅰ、 AMI-aphA1和AMIaadA1在16和24小时时两种制剂的结果差异有统计学意义( P<0.05)。其余时间段结果差异无统计学意义( P>0.05)。结论 蟾酥协同氨基糖苷类药物抗多重耐药鲍曼不动杆菌的机制与其抑制aac( 6') -II和aac( 6') -Ⅰ的表达有关,并且此抑制作用具有一定的时效性,醇提蟾酥对鲍曼不动杆菌氨基糖苷类药物耐药相关基因的表达抑制作用优于水提蟾酥。 |

| [Abstract] Objective To discuss the impacts of different times of Venenum Bufonis on the expression of aminoglycosides resistance gene. Method The impacts of different times of Venenum Bufonis (two extracting solution) on the expression of aminoglycosides resistance gene by RT-PCR.Result The expression of aac( 6') –II and aac( 6') -Ⅰwere decreased after 16 to 24 hours with Venenum Bufonis (alcohol extract and aqueous extract), which were significant lower than any other time(P<0.05), with statistical significance. Venenum Bufonis (alcohol extract ) has weak inhibition effects on the expression of ant(3〞)-Ⅰafter 16 hours. The inhibition effects of Venenum Bufonis (alcohol extract) on AMI-aphA1 and AMIaadA1 was coincident?with Venenum Bufonis (aqueous extract) for two hours. The CT value of acc(6’)-II, acc(6’)-I, ant(3’)-I, AMI-aphA1 and AMI-aphA1 were statistically significant difference between Venenum Bufonis(alcohol extract) and Venenum Bufonis(aqueous extract) at 16 o’clock and 24 o’clock(P<0.05). However, there were no significant difference at any other time(P>0.05). Conclusion Venenum bufonis(especially alcohol extract) and aminoglycosides against multi-drug resistant Acinetobacter Baumannii relative to the inhibition of transacetylase aac( 6') –II and aac( 6') -Ⅰ. |
