| 目的 观察不同浓度补阳还五汤(Buyanghuanwu decoction,BYHWD)含药血清对人脑微血管内皮细胞(human brain microvascular endothelial Cells,HBMECs)增殖和迁移影响。方法 补阳还五汤灌胃大鼠后腹主动脉取血,制备含药血清;体外培养人脑微血管内皮细胞(HBMECs),随机分为空白组(正常大鼠血清)及补阳还五汤含药血清组,血清浓度分为5%、10%和20% 3个浓度,共6组;用四甲基偶氮唑蓝(MTT)法检测HBMECs增殖;细胞划痕法检测HBMECs迁移的影响。结果 与空白组相比较,含药血清组显著促进HBMECs增殖和迁移(P<0.05);与10%含药血清组比较,5%和20%含药血清组细胞增殖和迁移的相对距离显著减小(P<0.05)。结论 补阳还五汤含药血清促进HBMECs增殖和迁移, 10%含药血清作用最佳。 |

| Objective To observe the different concentration of Buyanghuanwu decoction on human brain microvascular endothelial cells proliferationand migration. Methods HBMECs were randomly divided into blank group (normal rat serum) and Buyanghuanwu decoction drug-containing serum group, group divided into 5%, 10% and 20% three concentrations, total of six groups. After cultivated 24 h, the brain microvascular endothelial cell proliferation was determined by MTT,the influence of migration was detected by cell scratch method. Results Buyanghuanwu decoction concentration dependence in cell proliferation, the same point in time is compared between each group differences were statistically significant. Compared with blank group, drug-containing serum significantly promote cell proliferation and relative distance(P<0.05). Compared with blank group, the cell migration of 5% of BYHWD, 10% BYHWD and 20% BYHWD proliferation and relative distance increased significantly(P<0.05). Conclusion 10% BYHWD can significantly promote HBMECs proliferation and migration. |
