| [目的]研究益气养阴祛瘀中药对干燥综合征模型样NOD小鼠外周血单个核细胞TLR9、IFN-α、BAFFmRNA表达水平的调节作用,探讨益气养阴祛瘀中药对SS的TLR-IFN-BAFF通路信号表达的影响及其作用机制。[方法]实验动物选择NOD小鼠20只及ICR正常小鼠10只,雌性,8周龄,SPF级,于无特殊病菌的恒温条件下喂养。NOD小鼠随机分为2组:模型组、给药组。ICR正常小鼠10只,作为正常对照组。于小鼠9周龄时开始每日定时灌胃一次,持续12周。于实验第12周末时所有小鼠股动脉取血后处死,分离外周血单个核细胞,采用RT-PCR法检测TLR9、IFN-α、BAFF的mRNA表达水平。小鼠股动脉取血后处死,摘取小鼠颌下腺,用光镜下观察小鼠颌下腺病理改变。[结果] 对照组、模型组、中药组TLR9 mRNA表达水平分别为0.15±0.03,0.92±0.05,0.29±0.06;对照组、模型组、中药组IFN-α mRNA表达水平分别为0.12±0.03,1.48±0.04,0.20±0.03;对照组、模型组、中药组BAFF mRNA表达水平分别为0.23±0.06,1.28±0.04,0.38±0.06;模型组和正常对照组相比TLR9、IFN-α、BAFFmRNA均明显升高(P<0.01 或P<0.05),给药组和模型组比较TLR9、IFN-α、BAFFmRNA均明显下降(P<0.05)。[结论]NOD小鼠存在血清TLR9、IFN-α、BAFF mRNA水平过度表达。益气养阴祛瘀中药对NOD小鼠TLR/IFN/BAFF通路的过度表达具有抑制作用。 |

| [Objective]The aim of the study is,based on the effective applying strengthening qi, nourishing yin, quyu to treat Sjogren"s syndrome,through studying the impact on Spontaneous models, and the expression of TLR9, IFN-α, BAFF mRNA in Peripheral blood mononuclear cells(PBMC)of NOD mice to explore the possibility of using strengthening qi.nourishing yin.quyu to cure SS and the mechanism of the impact of expression of TLR-IFN-BAFF signal pathway of SS.[Methods]20 NOD mice and 10 normal are selected,all of which are eight weeks old,female, SPF level( Shang slac laboratory animal CO.LTD ), and are raised under the condition of constant temperature conditions without special bacteria.20 NOD mice were randomly divided as 2 groups: control group, TCM group. ICR group consist ten normal mice, which is the reference group. At the end of the twelveth week, the blood from all mice"s arteriae femoralis .was fetched. Mice were killed after its femoral artery"s blood is fetched. PBMC (Peripheral blood monocytes cell) were Isolated from peripheral blood and examined by reverse transcription-polymerase chain reaction (RT-PCR) analysis for mRNA expression of TLR9,IFN-α and BAFF. Mice were killed after its femoral artery"s blood is fetched,submandibular Glande were fetched to abserve their pathological changes under light microscopic.[Results] Compared to normal control group, the expression levels of TLR9,IFN-α and BAFF mRNA of NOD group were found to be increased(P <0.01); Compared to the model group, the expression level of TLR9,IFN-α and BAFF mRNA of the treatment groups were found to be reduced(P<0.05);.Conclusion:TLR9, IFN-α and BAFF levels in the serum was significantly increased in the NOD mice.Strengthening qi, nourishing yin,quyu significantly reduce TLR9, IFN-α, BAFF overexpression in serum. |
