| 目的 研究宫内暴露于七氟醚对仔鼠海马CA1区神经元凋亡及早期认知功能的影响。方法 成年性成熟wistar雌鼠交配妊娠后取孕鼠8只,随机分为对照组(C组),七氟醚组(S组),每组n=4。S组于孕19天时暴露于2.4%七氟醚6h,C组不给予任何药物。应用免疫组化法观察两组仔鼠在出生后第1天(Postnatal Day 1,PN1 )、第14天(PN14)、第21天(PN21)海马CA1区caspase-3的表达情况。出生后32天-37天对仔鼠进行Morris水迷宫实验以测定其认知功能。结果 1、出生后第1天, S组仔鼠海马CA1区caspase-3的总光密度值(Integral Optical Density,IOD)明显高于C组仔鼠[(60308.6±6403.6)比(17680.5±9326.7),P <0.05],出生后第14,21天,S组仔鼠海马CA1区caspase-3的总光密度值与C组仔鼠相比无统计学差异[(3213.7±582.8)比(8503.2±2402.5),(2046.0±62.7)比(4839.9±716.3),P >0.05]。2、S组仔鼠逃避潜伏期与C组仔鼠相比无统计学差异[(80.0±13.5)秒比(80.7±19.1)秒,(52.2±10.3)秒比(41.0±19.7)秒,(29.3±10.9)秒比(28.5±15.3)秒,(22.0±5.5)秒比(19.2±6.7)秒,(16.2±2.8)秒比(20.3±4.8)秒,P >0.05],S组仔鼠穿越目标象限次数与C组仔鼠相比无统计学差异[(8.3±0.8)次比(8.2±3.0)次,P >0.05]。结论 孕晚期Wistar母鼠暴露于2.4%七氟醚对仔鼠海马CA1区神经元发育的影响呈“一过性损伤”,并不会引起子代早期认知功能异常。 |

| Objective To investigate the effect of sevoflurane exposure in uterus on the Wistar rat offspring’s brain neural apoptosis in the CA1 region of hippocampus and the cognitive function at early age. Methods Eight adult female Wistar rats got pregnant. With the random number table method, pregnant rats were divided into two groups, control group(group C)and sevoflurane group(group S), n=4. The rats in group S exposed to 2.4% sevoflurane for 6 hours on the gestational 19th day, while those in group C lived in the same condition without sevoflurane. The expression of caspase-3 in the CA1 region of hippocampus on PN1、PN14 and PN21 were observed by immunohistochemistry. The early cognitive function of the offspring was tested by Morris water maze test from PN32 to PN37. Results 1、Compared to the offspring of group C, the offspring of group S had the higher value of Integral Optical Density (IOD) in the CA1 region of hippocampus on PN1 [(60308.6±6403.6) vs (17680.5±9326.7), P <0.05]. On PN14 and PN21, the value of IOD number in the CA1 region of hippocampus for the offspring of group S was similar to that of group C[(3213.7±582.8) vs (8503.2±2402.5), (2046.0±62.7) vs (4839.9±716.3), P >0.05] 2、There were no significant differences in escape latency, explore number in the target quadrant between the offspring of group S and C [(80.0±13.5)s vs (80.7±19.1)s, (52.2±10.3)s vs (41.0±19.7)s, (29.3±10.9)s vs (28.5±15.3)s, (22.0±5.5)s vs (19.2±6.7)s, (16.2±2.8)s vs (20.3±4.8)s ; (8.3±0.8)times vs (8.2±3.0)times, P >0.05]. Conclution The effect of brain neural development in the CA1 region of hippocampus for the offspring of those who exposed to 2.4% sevoflurane during late pregnancy present “temporary injury”. It has no significant effect on the cognitive function at the early age for the offspring. |
