| 目的 探究黄芪甲苷对大鼠骨骺干细胞的增殖及生物学特性的作用影响。方法 将新生SD大鼠离体培养获取大鼠骨骺干细胞,选择黄芪甲苷对第3代骨骺干细胞进行干预诱导,体外培养,在倒置显微镜下分别观察实验组和对照组的细胞生长情况、记录细胞贴壁情况及时间、细胞形态变化及增殖情况。采用免疫组化Ⅱ型胶原单克隆抗体染色方法,检测诱导后细胞的生物学特性,MTT法检测细胞增殖活性。结果 经黄芪甲苷(100ug/ml ,3ml/d)干预诱导后,在倒置显微镜下可观察到实验组细胞增殖速度加快(对照组细胞群体倍增时间为30.17h ,实验组为23.20h),Ⅱ型胶原单克隆抗体染色表达阳性(实验组强于对照组),第6天MTT法检测出实验组od值(0.612+0.151)明显高于对照组(0.517+0.153)。结论 黄芪甲苷具有促进骨骺干细胞的增殖作用。经黄芪甲苷诱导培养的骨骺干细胞可以正常地分泌特征性基质,并保持骨骺干细胞的生物学特性。 |

| Objective The research aim to investigate the effect on proliferation and biological characteristics by Astragaloside in Rats precartilaginous stem cells. Methods The experiment obtain epiphysis stem cells by the method of culture newborn SD rats in vitro, chosing astragaloside induce the third generation epiphysis stem cells in vitro culture. Observe growth of cells of experimental group and the control group by inverted microscope and record cells of adhere time and circumstances, morphologic changes of cells and proliferation. Using immunohistochemistry Collagen typeⅡ monoclonal antibody staining test cell biological characteristics which was induced by astragaloside, and using MTT test the activity of Epiphysis stem cells. Results It makes the speed of cell proliferation accelerate through inverted microscope(population doubling time of control group is 30.17h, experiment group is 23.20h) after induction with astragaloside(100ug/ml ,3ml/d). Immunohistochemistry Collagen typeⅡmonoclonal antibody staining is positive(experiment group higher than control group). Optical density of experiment group(0.612+0.151) is higher than control group(0.517+0.153) by MTT method testing on the sixth day. Conclusion This research proved that Astragaloside can promote growth of precartilaginous stem cells. Precartilaginous stem cells which was induced with astragaloside can secrete characteristic matrix normally and keep the well biological characteristics of precartilaginous stem cells. |
