      目的:通过观察华蟾素注射液对乳腺癌MDA-MB-231细胞E-cad、N-cad表达的影响,探讨华蟾素注射液对乳腺癌细胞上皮间质转化(EMT)的影响并初步探究华蟾素注射液抑制乳腺癌转移机制。方法:采用四氮唑蓝(MTT)比色法观察不同浓度华蟾素注射液(1:5、1:10、1:20、1:40、1:80、1:160、1:320稀释)对MDA-MB-231细胞增殖影响;实时荧光定量PCR(qRT-PCR)检测法检测华蟾素注射液处理MDA-MB-231细胞后,细胞中E-cad/N-cad mRNA表达水平;蛋白印迹法(Westernblot)检测华蟾素注射液处理MDA-MB-231细胞后,细胞中E-cad/N-cad蛋白表达水平。结果:高浓度华蟾素注射液48h对MDA-MB-231细胞抑制率较中剂量、低剂量组更加明显,差异具有统计学意义(P<0.05),实时荧光定量PCR结果显示华蟾素注射液可以下调MDA-MB-231细胞N-cad的表达和上调E-cad表达,与空白组相比,差异具有统计学意义(P<0.01)。Westernblot结果显示华蟾素注射液能降低N-cad蛋白的表达和上调E-cad蛋白的表达,与空白组相比,差异具有统计学意义(P<0.05)。结论:华蟾素能通过上调E-cad的表达和下调N-cad的表达,抑制MDA-MB-231细胞的上皮间质转化(EMT)转变,从而降低乳腺癌MDA-MB-231细胞的转移能力。

      [Purpose]To investigate the effect of Cinobufotalin Injection on the expressions of E-cad, N-cad in breast cancer a Cell Line MDA-MB-231, in order to explore the inhibiting effect of Cinobufotalin Injection on the metastasis of breast cancer.[Methods] Different concentrations of Cinobufotalin Injection were prepared(1:5、1:10、1:20、1:40、1:80、1:160、1:320 diluted), and proliferation effects on cell line lovo were observed by MTT.The expressions of E-cad and N-cad mRNA were detected by qRT-PCR, and the expression of E-cad and N-cad protein was detected by Western Blot assay.[Results] The high concentration group of Cinobufotalin Injection had a higher cell inhibition effect at 48 hours, compared with the medium group and low group (P<0.05).The expressions of N-cad in drug group were down-regulated and the expression of E-cad was up-regulated,Compared with the blank and normal control group by qRT-PCR (P<0.01) and Western Blot assay (P<0.05).[Conclusion]Cinobufotalin Injection can inhibit the EMT transition of cell line MDA-MB-231 and decrease its migration and invasive ability through the down-regulation of N-cad expressions and the up-regulation of E-cad expression.
