| 目的 观察洋金花总碱对TGF-β1诱导的A549 细胞E-cad表达的变化。方法 将体外培养的A549 细胞随机分成5 组：空白对照组,TGF-β1组及低、中、高浓度药物组,分别作用24、48、72小时后,通过Western-blot及RT-PCT检测各组A549细胞E-cad表达的变化。结果 不同浓度作用的洋金花总碱药物组能在蛋白水平和mRNA水平使E-cad表达上调(P<0.01),并且在72小时及高浓度时最明显(P<0.01)。结论 洋金花总碱能增加上皮标志物E-cad的表达,且呈浓度—时间依赖性,能一定程度上抑制A549细胞的上皮间质转化。 |

| Objective ：Observation the E-cad expression changes after Yangjinhua alkaloids effect the A549 cell of TGF-β1-induced. Methods：In vitro, A549 cells were randomly divided into 5 groups, including, Blank control group,TGF-β1 group, low、medium and high concentration of drug group, observating the change of the cell morphology and examination of the expression of E-cad by Western blot and RT-PCR after 24 , 48 and 72 hours respectively. Results : Effect of different concentrations of the drug group Yangjinhua alkaloids can make E-cad increase in the protein and mRNA levels (P <0.01),and the change was the most obvious in 72 hours and the high level of drug group (P < 0.01) .Conclusion : Yangjinhua alkaloids can increase the expression of the epithelial marker like E-cad, depending on the time -the concentration and it inhibition the EMT process of A549 cells. |
