| 目的 原花青素具有抗抑郁作用,本研究主要探讨原花青素对抗抑郁药度洛西汀在大鼠体内药动学的影响。方法 将15只健康雄性大鼠随机分为三组:对照组、单次给药组和多次给药组(各5只)。多次给药组连续灌胃给予100 mg.kg-1原花青素8天、单次给药组第8天灌胃给予100 mg.kg-1原花青素后,每组大鼠均灌胃给予度洛西汀40 mg.kg-1,并按时间点经大鼠尾静脉采集0.2-0.3 mL血样,采用HPLC法检测度洛西汀浓度。计算相应的药动学参数并进行统计学分析。结果 单次给予原花青素对度洛西汀的药动学参数无显著影响;多次给予原花青素后,仅度洛西汀的Tmax显著减小,其余参数未有显著变化。结论 作为一类有潜力的天然抗抑郁药,原花青素对度洛西汀的药动学影响小,其与度洛西汀合用治疗抑郁症有待进一步的研究。 |

| OBJECTIVE Procyanidins(PC) have antidepressant effect. We aimed to investigate the effect of procyanidins on the pharmacokinetics of duloxetine in rats. METHOD 15 healthy male rats were randomly divided into three groups: a control group, a single-dosage group and a multiple-dosage group. The multiple-dosage group received 100 mg.kg-1.d-1 procyanidins for 8 days and the single-dosage group received 100 mg.kg-1 procyanidins on the 8th day, followed by an oral administration of 40 mg.kg-1 duloxetine to all the rats. Blood samples were collected via tail vein at the given time points and analyzed for duloxetine concentration by HPLC. RESULT The pharmacokinetic parameters were calculated and analyzed statistically. No significant change was observed except Tmax of the multiple-dosage group. CONCLUSION As a potential antidepressant, procyanidins has modest effect on the pharmacokinetics of duloxetine. Coadministration of these two drugs to treat depression need further investigation. |
