| 目的:筛选出5-氟苄氧基杨梅醇抗人肺腺癌A549作用强的同分异构体。方法:通过3H-TdR掺入法观察5-对(间、邻)氟苄氧基杨梅醇体外对人肺腺癌A549细胞的抑制作用,确定抑瘤效果最好的5-氟苄氧基杨梅醇结构。结果:100、50、25、12.5、6.25、3.125 μg/ml?5-对(间、邻)氟苄氧基杨梅醇衍生物抑制率分别为?5-对氟苄氧基杨梅醇衍生物抑制率:(100.0±0.0)%、(100.0±0.0 )%、(99.5±0.8)%、(80.6±6.7)%、(42.0±5.5)%、(26.5±4.3)%;5-间氟苄氧基杨梅醇衍生物抑制率:(100.0±0.0)%、(99.9±0.0)%、(90.6±3.2)%、(49.2±8.5)%、(22.1±3.6)%、(0.7±1.0)%;5-邻氟苄氧基杨梅醇衍生物抑制率:(100.0±0.0)%、(100.0±0.0)%、 (92.5±4.2)%、(60.2±10.6)%、(10.7±2.3)%、(4.8±2.8)%。5-对(间、邻)氟苄氧基杨梅醇衍生物对A549细胞均有较好的抑制作用,随着浓度的降低,抑制作用减弱,其中5-对氟苄氧基杨梅醇抑瘤效果最好,在3.125μg/ml浓度下,也有较好的抑制作用。结论:5-对(间、邻)氟苄氧基杨梅醇衍生物对A549细胞均有较好的抑制作用,其中,5-对氟苄氧基杨梅效果最好。 |

| Objective:To screen the Most effective 5-fluorobenzyloxy Myricanol on Resisting A549 in vitro from three isomers. Methods: To determine the best anti-tumor effect, sensitivity test of 3H-TdR incorporation assay was used to detect the effect of inhibiting A549 cells by 5- p (m, o)-fluorobenzyloxy Myricanol in vitro.Results:?The proliferation inhibition rate?of A549 cells treated with?100, 50, 25, 12.5,6.25, 3.125 μg/ml?5-p(m,o)-fluorobenzyloxy Myricanol?was as follows: 5-p-zyloxy Myricanol : (100.0±0.0)%,(100.0±0.0 )%,(99.5±0.8)%,(80.6±6.7)%,(42.0±5.5)%,(26.5±4.3)%; 5-m-fluorobenzyloxyMyricanol:(100.0±0.0)%,(99.9±0.0)%,90.6±3.2)%,(49.2±8.5)%,(22.1±3.6)%,(0.7±1.0)%; 5-o-fluorobenzyloxy Myricanol:(100.0±0.0)%,(100.0±0.0)%,(92.5±4.2)%,(60.2±10.6)%,(10.7±2.3)%,(4.8±2.8)%.5-p (m, o)-fluorobenzyloxy Myricanol had good inhibition effect on A549 cell .The inhibition effect decrease with the reduction of the concentration. Among them, 5-p-fluorobenzyloxy Myricanol still have good inhibitory effect under the concentration of 3.125μg/ml. Conclution:5-p (m, o)-fluorobenzyloxy Myricanol derivative has good effect on resisting tumor. And among them , 5-p-fluorobenzyloxy Myricanol has the best effect. |
