      目的 探讨miRNA-20a-BCL-2信号通路介导雷公藤红素、平阳霉素对舌鳞癌细胞增殖与侵袭。方法Tca8113细胞液于DMEM培养液中培养,作为对照组;雷公藤红素组、平阳霉素组、联合组的培养方法同对照组,10%胎牛血清DMEM培养液分别含有20μg/ml的雷公藤红素、20μmol/ml的平阳霉素组以及20μg/ml雷公藤红素组+20μmol/ml 平阳霉素。以上各组每孔设6个平行样,培养72h,培养结束后,检测Tca8113细胞活力、凋亡、侵袭情况,实时荧光逆转录法测定miRNA-20a、BCL-2 mRNA水平,酶联免疫吸附法测定MMP-9、cyc D1、VEGF蛋白水平。结果 雷公藤红素组、平阳霉素组、联合组OD值、miRNA-20a 、BCL-2 RNA、Tca8113穿膜数、MMP-9、cyc D1、VEGF表达水平均低于对照组,抑制率、Tca8113细胞凋亡率高于对照组(P<0.05);联合组OD值、miRNA-20a 、BCL-2 RNA、Tca8113穿膜数、MMP-9、cyc D1、VEGF低于雷公藤红素组、平阳霉素组,抑制率、Tca8113细胞凋亡率高于雷公藤红素组、平阳霉素组(P<0.05);雷公藤红素组OD值、存活率、miRNA-20a 、BCL-2 RNA、Tca8113穿膜数、MMP-9、cyc D1、VEGF、Tca8113细胞凋亡率与平阳霉素组相近(P>0.05)。结论 雷公藤红素联合平阳霉素能抑制miRNA-20a-BCL-2信号通路及下游细胞因子的表达,进而抑制舌鳞癌细胞增殖、侵袭。

      Objective To investigate the role of miRNA-20a-BCL-2 signaling pathway in the proliferation and invasion of tongue squamous cell carcinoma by triptolide and pingyangmycin. Methods Ca8113 cell solution (cell concentration 5×106/mL) was cultured in DMEM medium containing 10% fetal bovine serum in a CO2 incubator (37°C, 5% CO2, 20% O2) as a control group: Leigong The culture method of the lignin group, the pingyangmycin group and the combination group was the same as that of the control group, and the 10% fetal bovine serum DMEM medium contained 20 μg/ml tripterine, 20 μmol/ml pingyangmycin group and 20 μg/ml, respectively. Tripterine group +20 μmol/ml pingyangmycin. Six replicates per well in each group were cultured for 72 h. After the culture, the viability, apoptosis and invasion of Tca8113 cells were detected. The mRNA levels of miRNA-20a and BCL-2 were determined by real-time fluorescence reverse transcription. MMP was determined by enzyme-linked immunosorbent assay. -9, cyc D1, VEGF protein levels. Results The OD value, survival rate, miRNA-20a, BCL-2 RNA, Tca8113 transmembrane number, MMP-9, cyc D1, VEGF expression levels of tripterine group, pingyangmycin group, combined group were lower than that of control group (P<0.05). The combined group OD value, survival rate, miRNA-20a, BCL-2 RNA, Tca8113 transmembrane number, MMP-9, cyc D1, VEGF were lower than Leigong. The apoptosis rate of Tca8113 cells in the lignin group and the pingyangmycin group was higher than that in the tripterine group and the pingyangmycin group (P<0.05). The OD value and survival rate of the tripterine group were the same. The apoptosis rates of miRNA-20a, BCL-2 RNA, Tca8113 transmembrane, MMP-9, cyc D1, VEGF and Tca8113 cells were similar to those of pingyangmycin group Conclusion Tripterine combined with pingyangmycin can inhibit the expression of miRNA-20a-BCL-2 signaling pathway and downstream cytokines, and inhibit the proliferation and invasion of tongue squamous cell carcinoma.
