      目的明确七叶皂苷钠(ESC)对糖尿病大鼠血-视网膜屏障(BRB)的保护作用,进一步探讨其作用机制。方法采用链脲佐菌素(STZ)诱导SD(Sprague Dawley)大鼠建立糖尿病性视网膜病变(DR)模型,予以七叶皂苷钠(1.5mg/kg?d)灌胃处理,通过检测视网膜中伊文思蓝(EB)渗漏量评估ESC对DR大鼠BRB的影响;采用Western blot法检测视网膜组织Occludin蛋白、Rho相关卷曲螺旋形成蛋白激酶(Rho associated coiledcoil forming protein kinase,ROCK)蛋白及磷酸化肌球蛋白轻链(P-MLC)蛋白表达的量。结果ESC能够减少DR大鼠的BRB渗漏情况(P < 0.01),ESC能够增加Occludin蛋白表达(P < 0.01),同时抑制ROCK/P-MLC通路的激活(P < 0.01)。结论七叶皂苷钠对糖尿病大鼠BRB具保护作用,其机制为增加DR大鼠视网膜组织Occludin蛋白的表达,同时抑制炎性信号通路的激活。
英文摘要:
      Objective:To testify the protection effects of ESC on blood-retinal barrier (BRB) in diabetic rats, and investigate its mechanism.Methods:DR modelwas induced by STZ injection in SDrats and then the DR rats were given with 15 mg/kg?dESC through the stomach. The effect of ESC to BRB was evaluated by the expression of Evansblue(EB) leakage in retial;Thelevles of Occludin,ROCK and its substrate P-MLCin retialwere detected by Western blotting. Results:TheEB leakage through BRBwas reduced by ESC(P < 0.01)which increase the expression of Occludin(P < 0.01)and also inhibitethe activation of ROCK pathway(P < 0.01).Conclusion:ESCcan protect BRB of DR rats by increasing the expression of Occludinin retial of DR rats and inhibiting the activation of the inflammatory signaling pathway.
