| 目的 建立去甲斑蝥素脂质体的包封率测定方法。方法 采取高速离心法分离脂质体和游离去甲斑蝥素,采用HPLC法测定去甲斑蝥素的含量,计算脂质体中去甲斑蝥素的包封率;色谱条件:采用Agilent C18(150 mm×4.6 mm,4μm)色谱柱;以乙腈-磷酸水溶液(pH3.1)(10∶90)为流动相,流速为0.8 mL.min-1;检测波长210nm;柱温为25℃。结果 去甲斑蝥素与脂质体辅料的色谱峰分离良好,去甲斑蝥素线性范围9.18μg.mL-1~459.00μg.mL-1(r=0.999),高中低浓度的加样回收率范围为98.87%~100.31%,包封率为25.53%。结论 该方法适用于检测去甲斑蝥素脂质体中去甲斑蝥素的含量及包封率。 |

| Objective To establish a method for the determination of entrapment efficiency of norcantharidin liposomes. Methods The content of norcantharidin in liposomes was determined by high-speed centrifugation and the entrapment efficiency of norcantharidin in liposomes was calculated by HPLC. The chromatographic conditions were as follows:Agilent C18 column (150 mm x 4.6 mm, 4μm). The flow rate was 0.8 mL.min-1.The detection wavelength was set at 210nm and the column temperature was 25℃. Results The chromatographic peaks of norcantharidin and liposome excipients were well separated. The linear range of norcantharidin was 9.18μg.mL-1~459.00μg.mL-1 (r = 0.999).The recovery ranged from 98.87% to 100.31% at high and low concentrations, and the entrapment efficiency was 25.53%. Conclusion The method is suitable for the detection of norcantharidin in norcantharidin liposomes and its entrapment efficiency. |
