| 目的 探讨自然铜、鹿衔草在裸鼠肺癌骨转移过程中对Intercellular cell adhesion molecule-1 (ICAM-1,细胞间粘附分子-1)、Matrix metalloprotein-9(MMP-9,基质金属蛋白酶-9)及Parathyroid hormone-related peptide(PTHrP,甲状旁腺激素相关蛋白)表达水平的影响。方法 以A549肺癌细胞建立肺癌胫骨局部骨转移模型。将模型动物随机分为5组(自然铜组,鹿衔草组,中药合用组,阳性对照组及空白对照组)。中药组裸鼠给与中药灌胃,阳性对照组腹腔注射帕米磷酸二钠注射液;空白对照组给与对应生理盐水处理。3周后收集肿瘤标本,采用免疫组织化学法及实时荧光定量PCR法(qRT-PCR)检测ICAM-1、MMP-9、PTHrP的蛋白表达水平及基因表达量。结果 自然铜组,鹿衔草组及中药合用组表达ICAM-1的阳性细胞百分率分别为(35.2±1.1)%、(33.6±4.0)%、(38.3±4.6) %,表达MMP-9的阳性细胞百分率为(59.2±0.7) %、(65.4±1.7) %、(67.7±1.7) %,均低于空白对照组(P <0.01)及阳性对照组(P<0.05);中药合用组表达PTHrP的阳性细胞百分率为(73.3±7.2) %, 低于空白对照组(P <0.05)。自然铜、鹿衔草及中药合用组ICAM-1的基因相对表达量分别为(10±0.5) ×10^-2、(7.3±0.6) ×10^-2、(6.6±1.1) ×10^-2,自然铜、鹿衔草及中药合用组MMP-9的基因相对表达量分别为(11.9±2.2) ×10^-2、(21.7±5.1) ×10^-2、(23.4±5.9) ×10^-2,均低于空白对照组(P <0.01)及阳性对照组(P<0.05);中药合用组PTHrP的基因相对表达量为(74.7±17.1) ×10^-2,低于空白对照组水平(P <0.01)结论 自然铜、鹿衔草及其联合用药通过下调ICAM-1、MMP-9、PTHrP的表达水平抑制裸鼠肺癌骨转移过程。 |

| Objective To investigate the effects of pyrite and herb pyrolae on ICAM-1, MMP-9 and PTHrP in bone metastasis of lung cancer in nude mice. Methods The model of bone metastasis of lung cancer was established by injected A549 cell suspension into marrow cavity of right tibia of nude mice. Then mice were randomly divided into pyrite group, herb pyrolae group, mixed drugs group (drugs are mixed with pyrite and herb pyrolae), pamidronate group and control group. Mice in pyrite group, herb pyrolae group and mixed drug group were given by gavage with pyrite, herb pyrolae and mixed drug by 6 g?kg-1every other day and injected 0.16mL normalSsalineSintraperitoneally every week, separately. Mice in pamidronate group was injected pamidronate disodium by 10 mg?kg-1 intraperitoneally every week and given by gavage with 0.5 mL normal saline every other day. Mice in control group was given by gavage with 0.5 mL normalSsaline every other day andinjected 0.16 mL normalSsalineSintraperitoneally every week. After 3 weeks, tumor specimens were collected. The expression of ICAM-1, MMP-9 and PTHrP were tested via immunohistochemistry and Real-time fluorescent quantitative PCR both at protein and gene level. Results when compared with control group, the expressions of ICAM-1 and MMP-9 in pyrite group, herb pyrolae group and mixed drugs group reduced (p<0.01);the expressions of PTHrP in mixed drug group and pamidronate group reduce (p<0.05). As compared to pamidronate group, the expressions of ICAM-1 and MMP-9 in Pyrite group, herb pyrolae group and mixed drug group reduced (p<0.05);the expressions of PTHrP in Pyrite group, Herb pyrolae group and mixed drug group rose(p<0.01). Conclusion Pyrite, herb pyrolae and their mixture could inhibit the expression of ICAM-1, MMP-9 and PTHrP during the bone metastasis of lung cancer in nude mice. |
