| 目的:观察制狗脊提取物对大鼠骨质疏松性骨折愈合的影响,同时初步探索其机制。方法:将32只SPF级SD雌性大鼠随机分成对照组、观察组,并依次造模。骨折造模24h后,对照组予以七厘散胶囊药液灌胃,观察组予制狗脊提取物灌胃,1次/日,分别干预2周、6周后腹主动脉取血分离血清检测钙磷水平、转化生长因子-β1(TGF-β1)与白介素-1β(IL-1β)水平,并收取骨折标本,固定后分别行骨密度检测、Micro-CT扫描。结果:骨质疏松造模6周后,对照组与观察组大鼠骨密度均低于正常水平;在干预2周、6周后观察组骨密度均高于对照组(P<0.05)。Micro-CT扫描显示观察组BV/TV、Tb.N、Tb.Th、SMI在干预后优于观察组,虽然BV/TV 、Tb.Th参数在干预后2周时间点差异无统计学意义。干预2周、6周后,比较组间钙磷水平、TGF-β1、IL-1β水平发现观察组钙磷水平在2周时间点显著高于对照组(P<0.05);在TGF-β1水平方面,观察组也均高于对照组(P<0.05);而在IL-1β水平方面,对照组IL-1β水平均高于观察组,且组间存在统计学意义差异。结论:本研究表明制狗脊提取物可增强骨质疏松骨折大鼠骨痂骨强度,改善骨丢失,促进骨痂生成与塑形,进而促进骨质疏松性骨折愈合,且其机制可能与其对TGF-β1、IL-1β水平分泌调节有关。 |

| ABSTRACT Objective: To explore the promoting effect Rhizoma Cibotii extract on osteoporotic rat fracture healing by the way of Micro-CT, X-ray, ELISA. Methods: 32 female SD rats were used in this experiment, all of them were randomly divided into control group and observation group, then base on 2w, 6w observation time point, rats were divided into another two. All were given the bilateral-ovariectomy, a unilateral open tibiae transverse fracture and fixed by intramedullary needle, successively. The observation group and control group were given Rhizoma Cibotii extract and Qili capsule respectively. After the operation, X-ray was used to make sure the fracture model is qualified, only the qualified one can be divided into the group. The characteristics of volume of callus (BV), bone volume of ROI (TV), bone volume fraction (BV/TV) and another related indexes from Micro-CT. Results: 6 weeks after ovariectomy, the bone density of rat decrsased than the normal. At 2w and 6w time point, the bone density in observation group was higher than it in control group(0.24±0.02,0.22±0.01;0.27±0.02,0.24±0.02(P<0.05)).There were statistical differences in BV/TV, Tb.Th, Tb.N, SMI between treatment group and control group at the time of 2w, 6w, though BV/TV, Tb.Th have no statistical differences at the first time point. At 2w,6w time point, there was statistical difference in serum levels of Ca2+, TGF-β1, and IL-1β between 2 groups(P<0.05), the level of Ca2+and TGF-β1 in observation group were higher than the other, but the IL-1β level was opposite(323.87±16.12,299.03±7.06;145.66±21.87,116.59±6.98).Conclusion: Rhizoma Cibotii extract has promoting effect on osteoporotic fracture healing of rats, increase bone density and improve bone microstructure through influence the level of TGF-β1 and IL-1β.
