| 目的:挖掘中医药治疗骨肿瘤的用药规律,并基于网络药理学探讨高频药物治疗骨肿瘤的药理机制。方法:检索中国知网、万方数据知识服务平台、维普网中的文献,使用Excel 2016,SPSS Modeler 18.0,SPSS Statistics 24.0软件,将纳入文献的中药进行聚类分析、关联规则分析,从DisGENT数据库及TTD数据库提取骨肿瘤的主要靶点,TCMSP数据库中提取高频药物的主要成分,通过ChemMapper数据库提取主要成分的预测靶点,使用Cytoscape 3.2.1软件构建成分-靶点网络,进一步分析主要通路及潜在作用机制。结果:最高频次用药为黄芪,使用最多的药物类别为补虚药,药物四气五味主要以甘、温为主,归经中肝经最多,关联规则强度最高的组合有6对,聚类分析可归为补肾填精、化痰利湿散结、补益气血3类。桦木酸为黄芪主要发挥作用的成分,GSK-3β为核心靶点,主要作用于PI3K/AKT信号通路。结论:骨肿瘤的治疗多以黄芪为主药,可随证配伍补肾填精、活血化瘀、益气养血等药物,可能通过PI3K/AKT信号通路发挥作用,为后续基础及临床试验奠定基础。 |

| Objective: To explore the medication rules of Traditional Chinese Medicine(TCM) in treatment of primary bone tumor, and analyze the pharmacological mechanism of high frequency drugs based on network pharmacology. Methods: Literatures in CNKI, Wanfang and Weipu Network were retrieved. Excel 2016, SPSS Modeler 18.0, SPSS Statistics 24.0 software were used to analyze for clustering and association rules of the medicine. The main targets of bone tumors were extracted from Dis database and TTD database, while the main components of high-frequency drug were extracted from the TCMSP database. Then extract the main target of active ingredients form ChemMapper, and use Cytoscape 3.2.1 software to build composition-target network, so as to analysis the major pathway and potential mechanisms. Results: The most frequent medication was Radix Astragali. The most commonly used medicines was deficiency-nourishing herbs. The four natures and five flavors of the drug were mainly sweet and warm, and the major meridian tropism of TCM was liver meridian. There were 6 core medication combinations through association rules, while cluster analysis could be classified into three categories: tonifying kidney and replenishing essence, resolving phlegm, dispersing stasis and removing dampness, replenishing qi and blood. Betulinic acid was the main component of Radix Astragali, and GSK-3β was the core target, which mainly played a role on the PI3K/AKT signaling pathway. Conclusion: Radix Astragali is the main drug for the treatment of bone tumors, and it can be combined with herbs as tonifying kidney and replenishing essence, activating blood circulation to dissipate blood stasis, replenishing qi and blood, which may play a role through the PI3K/AKT signal pathway, and laid the foundation for subsequent basic and clinical trial. |
