| 目的:研究二甲双胍对H2O2诱导的PC12 细胞损伤可能的保护作用机制。方法:使用CCK-8法检测二甲双胍对PC12细胞的细胞活力;建立H2O2损伤PC12细胞的细胞模型,使用CCK-8法和LDH法检测二甲双胍对PC12细胞生存率和LDH释放量,采用Hoechst 33342染色法和流式细胞术检测二甲双胍对PC12细胞的凋亡和ROS水平情况。qPCR和Western blot法检测二甲双胍对Nrf2/HO-1通路mRNA和蛋白表达水平。使用小干扰RNA沉默Nrf2检测二甲双胍对H2O2诱导细胞的保护作用。结果:二甲双胍在浓度0.5、1和2 mmol/L下对PC12细胞不表现毒性作用。预处理二甲双胍可对H2O2造成的PC12细胞损伤产生浓度依赖性保护作用和降低LDH。同时,二甲双胍降低H2O2造成的细胞凋亡和ROS水平。二甲双胍能激活细胞内Nrf2/HO-1通路起到抗氧化保护作用。结论:二甲双胍预处理后通过激活细胞内Nrf2/HO-1通路对H2O2诱导的 PC12 细胞损伤具有保护作用。 |

| Objective: To study the effect of metformin on PC12 cell injury induced by H2O2 and its possible molecular mechanisms.Methods: CCK-8 method was used to detect the cell viability of metformin on PC12 cells. The cell model of H2O2-induced PC12 cell damage was established, and the CCK-8 method and LDH method were used to detect the survival rate and the amount of LDH released, respectively. Hoechst 33342 staining method and flow cytometry were applied to detect the apoptosis and ROS level of PC12 cells. The expression levels of metformin on the mRNA and protein of Nrf2/HO-1were detect via qPCR and Western blot method. Use small interfering RNA to silence Nrf2 to detect the protective effect of metformin on H2O2-induced cells. Results: Metformin showed no toxicity to PC12 cells at concentrations of 0.5, 1 and 2 mmol/L. Pretreatment of metformin could produce a concentration- dependent protective effect on PC12 cell injury induced by H2O2 and reduce LDH. At the same time, metformin reduced cell apoptosis and ROS levels caused by H2O2. Metformin can activate the intracellular Nrf2/HO-1 pathway to play a role in antioxidant protection. Conclusion: Metformin pretreatment can protect PC12 cells injury caused by H2O2 through activating the intracellular Nrf2/HO-1 pathway. |
