      目的 研究槲皮素的分子特性,预测并验证槲皮素抗非小细胞肺癌(NSCLC)的潜在靶点。方法 TCMSP数据库分析槲皮素的药理学参数和分子特性;CTD和Swiss TargetPrediction预测槲皮素药物靶点;TTD分析NSCLC疾病靶点;分子对接筛选最佳靶点;蛋白免疫印迹实验验证靶点有效性。结果 槲皮素具有良好的成药性;通过多个数据库取交集和分子对接技术发现候选靶点--周期蛋白依赖性激酶1(CDK1);细胞实验表明槲皮素能有效减弱NSCLC细胞中CDK1的表达。结论 槲皮素可能通过降低细胞CDK1表达水平发挥抗NSCLC作用。
英文摘要:
      Objective To study the molecular characteristics of quercetin and screen the potential targets of quercetin against NSCLC. Methods TCMSP database was used to analyze the pharmacological parameters and molecular characteristics of quercetin. CTD and Swiss targetprediction were used to predict targets of quercetin. And targets of NSCLC were analyzed by TTD. The best target was screened by molecular docking, and western blot was used to verify the effectiveness of the target. Results Quercetin has good druggability. The candidate target cyclin dependent kinases 1 (CDK1) was found by multiple databases and molecular docking techniques. Cell experiments showed that quercetin could effectively reduce the expression of CDK1 in NSCLC cells. Conclusion Quercetin may play an anti-NSCLC activity by reducing the expression of CDK1.
