| 目的:癌性恶病质是一种以厌食、肌肉萎缩和体重减轻为特征的多因素综合征,不仅影响治疗,还降低了患者的生活质量,缩短了生存期。四君子汤是健脾益气的代表方,其适应症与癌性恶病质的症状相似,因此被广泛应用于治疗癌性恶病质。本研究旨在阐明四君子汤治疗癌性恶病质的机制。
方法:本研究采用一个全面的网络药理学策略,由四个顺序模块组成:目标预测,网络构建,分子对接,GO和KEGG富集分析。
结果:四君子汤中的7,9(11)-dehydropachymic acid和Taraxerol化合物可能通过作用于INS,PCK2,FABP3,eIF4E,SKP1,CCND1,CTNNB1,STAT5A,IL6ST,AR,CYCS,VEGFA等靶点来调节PI3K-Akt信号通路和Wnt信号通路,发挥调节代谢和抑制肿瘤的作用,最终达到治疗癌性恶病质的目的。
结论:四君子汤通过多个化合物、多个靶点、多个通路的直接或间接协同作用,能够有效地治疗癌性恶病质。 |

| Objective: Cancer cachexia (CC) is a multifactorial syndrome characterized by anorexia, weight loss, and muscle atrophy that affects not only treatment, but also lowers quality of life and shortens survival. Sijunzi Decoction (SJZD) is a representative prescription of traditional Chinese medicine (TCM) for strengthening spleen and replenishing qi. Its indications are similar to the symptoms of CC. And it is widely used in the treatment of CC. This study aims to clarify the mechanism of SJZD on CC.
Methods: This study used a comprehensive network pharmacology strategy consisting of four sequential modules: target prediction, network construction, molecular docking, GO and KEGG enrichment analysis.
Results: Compounds such as 7,9(11)-dehydropachymic acid and Taraxerol in SJZD may regulate PI3K-Akt signaling pathway and Wnt signaling pathway by acting on targets such as INS, PCK2, FABP3, eIF4E, SKP1, CCND1, CTNNB1, STAT5A, IL6ST, AR, CYCS, VEGFA and so on, so as to play a role in metabolic regulation and tumor inhibition, and finally achieve the goal of treating CC.
Conclusion: SJZD effectively acts against CC through the direct or indirect synergy of multiple compounds, multiple targets, and multiple pathways. |
