      目的 探究基因P4HA3在结直肠癌(colorectal cancer,CRC)的表达及与不良预后的关系。方法 从TCGA数据库获取CRC基因表达数据及临床特征,R软件对P4HA3表达及配对数据可视化分析。卡方检验P4HA3表达与CRC患者临床特征关系。R软件survival包对P4HA3进行生存分析。利用基因表达谱交互分析(GEPIA)数据库验证The Cancer Genome Atlas(TCGA)数据库CRC的P4HA3表达。通过GSEA软件对P4HA3进行KEGG通路富集分析?采用免疫组化染色检测30例CRC患者手术切除样本和30例癌旁组织样本中的P4HA3蛋白表达。结果 与对照组相比,P4HA3在CRC中高表达(P <0.01)。卡方检验显示P4HA3表达与CRC患者Stage、T和N分期密切相关(P <0.05)。生存分析显示与低表达P4HA3患者相比,P4HA3高表达CRC患者的生存时间明显缩短(P < 0.05)。通过GEPIA分析验证了基因P4HA3与CRC不良预后相关(P < 0.05)。进一步GSEA通路富集分析结果显示基因P4HA3富集到的信号通路与多种恶性肿瘤的发生发展有关(FDR < 0.05)。最后,免疫组化检测显示P4HA3蛋白在CRC中高表达,表明P4HA3在CRC发生发展中发挥作用。结论 高表达的基因P4HA3与CRC的进展及不良预后相关,其作用机制可能通过调控多种信号通路来实现。
英文摘要:
      Objective To explore the expression of gene P4HA3 in colorectal cancer and its relationship with adverse prognosis. Methods The gene expression data and clinicopathological data of colorectal cancer were obtained from TCGA database, and the gene P4HA3 expression data and paired data were visualized by R software. The relationship between the expression of P4HA3 gene and clinicopathological data of colorectal cancer patients was studied by chi-square test. The survival analysis of P4HA3 gene was carried out by using the survival package of R software. The expression of colorectal cancer gene P4HA3 in TCGA database was verified by gene expression profile interaction analysis (GEPIA) database. The KEGG pathway enrichment analysis of gene P4HA3 was performed by GSEA software. The expression of P4HA3 protein in surgical resection samples of 30 patients with colorectal cancer and adjacent tissues of 30 patients was detected by immunohistochemistry. Results Compared with control group, the gene P4HA3 was highly expressed in colorectal cancer tissues (P <0.01). Chi-square test showed that the expression of P4HA3 gene was correlated with stage, T and N stages of colorectal cancer patients (P <0.05). Survival analysis showed that compared with patients with low expression of P4HA3, survival time of colorectal cancer patients with high P4HA3 expression was significantly shortened (P <0.05). GEPIA analysis verified that gene P4HA3 was associated with poor prognosis of colorectal cancer (P <0.05). Further GSEA pathway enrichment analysis showed that the signal pathway enriched by gene P4HA3, and these was related to the occurrence and development of various malignant tumors (FDR < 0.05). Finally, immunohistochemical detection showed that P4HA3 protein was highly expressed in colorectal cancer tissues, which indicated the role of gene P4HA3 in the occurrence and development of colorectal cancer. Conclusion Highly expressed gene P4HA3 is associated with the progression and poor prognosis of colorectal cancer, and its mechanism may be achieved by regulating multiple signaling pathways.
