      目的:采用网络药理学方法及实验研究探讨复方刺梨合剂药抗胃癌的作用机制。方法:通过检索TCMSP数据库,获得复方刺梨合剂主要活性成分和相关作用靶点,通过CTD和OMIM数据库中筛选抗胃癌靶点,利用Cytoscape 3.7.1软件构建药物-成分-靶点网络图,获取蛋白质相互作用关系,绘制蛋白关系网络,采用WebGestalt和DAVID数据库进行GO功能和KEGG通路分析,采用Western Blot法测定p53、Bcl-2蛋白表达。结果:共筛选了复方刺梨合剂10个活性化学成分和236个化合物作用靶点,得到440个胃癌相关靶点和38个靶标,GO功能涉及对刺激反应、生物调节、细胞传递等,KEGG通路分析涉及63条显著信号通路,可见复方刺梨合剂通过参与调控多种生物学过程而发挥抗癌作用,实验结果证实复方刺梨合剂使用后p53和Bcl-2蛋白表达增加。结论:基于网络药理学方法及实验研究,揭示了复方刺梨合剂抗胃癌作用是多成分、多靶点、多通路作用的结果,可能通过调控p53和Bcl-2蛋白的表达发挥作用。
英文摘要:
      Objective To study mechanism and experimental of compound Cili mixture on gastric cancer based on network pharmacology. Methods The main active components and related targets of compound Cili mixture were obtained through the retrieval of TCMSP,the anti gastric cancer targets were selected through CTD and OMIM databases,the software of Cycloscape 3.7.1 was used to construct its network diagram of drug,disease and targets, protein interaction relationship was obtained and protein relationship network was drew,WebGestalt and David databases were used to analysis GO function and KEGG pathway,the protein expression of p53 and Bcl-2 was detected by Western Blot. Results A total of 10 main active chemical components of compound Cili mixture and 236 compound targets were screened,440 gastric cancer related targets and 38 targets were obtained,Go function involved response to stimulation,biological regulation,cell communication etc, KEGG pathway analysis involved 63 significant signaling pathways, it could be seen that compound Cili mixture played an anti-cancer role by participating in the regulation of various biological processes.The results showed that the protein expression of p53 and Bcl-2 increased after using compound Cili mixture. Conclusion It was revealed that the anti gastric cancer effect of compound Cili mixture was the result of multi-component,multi-target and multi-channel effect based on network pharmacology and experimental study, which may play a role by regulating the expression of p53 and bcl-2 protein.
