      目的 本研究旨在探讨槲皮素联合贯叶金丝桃提取物对脂多糖诱导小鼠抑郁样行为的影响及相关机制。方法 将90只大鼠随机分为采用随机数字表法分为槲皮素组、贯叶金丝桃提取物组、槲皮素联合贯叶金丝桃提取物组、盐酸氟西汀、模型组、对照组各15只。除对照组外,其余各组大鼠均单次注射1mg/kg脂多糖,将其制成抑郁模型。模型组不给予任何药物处理,槲皮素组给予50mg/kg槲皮素灌胃,贯叶金丝桃提取物组给予50mg/kg贯叶金丝桃提取物灌胃,槲皮素联合贯叶金丝桃提取物组给予槲皮素联合贯叶金丝桃提取物各50mg/kg灌胃,盐酸氟西汀给予2.6mg/kg盐酸氟西汀灌胃。对比各组糖水偏好、水迷宫实验结果、脑组织中炎症细胞因子水平及单胺类神经递质水平。结果 对照组及槲皮素联合贯叶金丝桃提取物组糖水摄入量、所占目标象限时间百分比、IL-10、DA、NA水平明显高于模型组、槲皮素组、盐酸氟西汀组、贯叶金丝桃提取物组,槲皮素组、盐酸氟西汀组、贯叶金丝桃提取物组糖水摄入量、所占目标象限时间百分比、IL-10、DA、NA水平均高于模型组,贯叶金丝桃提取物组糖水摄入量、所占目标象限时间百分比、IL-10、DA、NA水平明显高于槲皮素组、盐酸氟西汀组,差异有统计学意义(P<0.05)。对照组及槲皮素联合贯叶金丝桃提取物组潜伏期、IL-6水平明显低于模型组、槲皮素组、盐酸氟西汀组、贯叶金丝桃提取物组,槲皮素组、盐酸氟西汀组、贯叶金丝桃提取物组潜伏期、IL-6水平明显低于模型组,贯叶金丝桃提取物组潜伏期、IL-6水平明显低于槲皮素组、盐酸氟西汀组,差异有统计学意义(P<0.05)。结论 槲皮素联合贯叶金丝桃提取物可能通过减低炎症细胞因子水平,增加单胺类神经递质水平发挥抗抑郁作用。

      Objective The purpose of this study was to investigate the effect of quercetin combined with Hypericum perforatum extract on lipopolysaccharide-induced depression-like behavior in mice and related mechanisms. MethodsNinety rats were randomly divided into quercetin group, hypericum perforatum extract group, quercetin combined with hypericum perforatum extract group, fluoxetine hydrochloride, model group, and control group. 15 pieces. Except for the control group, rats in the other groups were given a single injection of 1 mg/kg lipopolysaccharide to make a depression model. The model group was given no drug treatment. The quercetin group was given 50mg/kg quercetin by gavage, the hypericum perforatum extract group was given 50mg/kg hypericum perforatum extract by gavage, and the quercetin combined with hypericum perforatum extract group was given by gavage. They were given quercetin combined with Hypericum perforatum extract 50mg/kg each by gavage, and fluoxetine hydrochloride was given by gavage 2.6mg/kg fluoxetine hydrochloride. Compare the sugar and water preferences, the results of the water maze experiment, the levels of inflammatory cytokines in the brain tissues and the levels of monoamine neurotransmitters in each group.ResultsIn the control group and the quercetin combined with hypericum perforatum extract group, the intake of sugar and water, the percentage of time in the target quadrant, the levels of IL-10, DA, and NA were significantly higher than those in the model group, quercetin group, and fluoxetine hydrochloride group , Hypericum perforatum extract group, quercetin group, fluoxetine hydrochloride group, Hypericum perforatum extract group sugar water intake, percentage of target quadrant time, IL-10, DA, NA levels are higher than the model group , Hypericum perforatum extract group sugar water intake, percentage of target quadrant time, IL-10, DA, NA levels were significantly higher than quercetin group, fluoxetine hydrochloride group, the difference was statistically significant (P<0.05 ). The incubation period and IL-6 levels of the control group and the quercetin combined with hypericum perforatum extract group were significantly lower than the model group, quercetin group, fluoxetine hydrochloride group, hypericum perforatum extract group, quercetin group, fluoride hydrochloride The incubation period and IL-6 levels of the oxetine group and the hypericum perforatum extract group were significantly lower than the model group, and the incubation period and IL-6 levels of the hypericum perforatum extract group were significantly lower than that of the quercetin group and the fluoxetine hydrochloride group. The differences were as follows: Statistically significant (P<0.05). Conclusion Quercetin combined with Hypericum perforatum extract may exert an antidepressant effect by reducing the level of inflammatory cytokines and increasing the level of monoamine neurotransmitters.
