| 目的 探讨白芨水煎剂对蒽醌类药物所致结肠黑变病(melanosis coli,MC)豚鼠结肠粘膜超微结构的影响。方法 将40只清洁级豚鼠按随机数字表法分为正常组(8只)、MC模型组(16只)及白芨组(16只)。MC模型组及白芨组均予大黄提取液6g/kg·d连续灌胃60d。造模开始2周后MC模型组予生理盐水灌肠,白芨组予10%白芨水煎剂灌肠,每周3次。观察各组豚鼠结肠上皮细胞褪黑色素染色、超微结构改变和流式细胞检测。结果 与正常组及白芨组相比,模型组豚鼠褪黑色素染色呈阳性改变,且范围明显较白芨组大;模型组电镜下可见多数上皮细胞胞浆内大小不等空泡样改变,偶可见凋亡上皮细胞,而白芨组个别线粒体轻度空泡样改变,上皮细胞凋亡不明显;与正常组(13.73±2.73%)相比较,模型组豚鼠的结肠上皮细胞凋亡率(35.27±1.21)增高(p<0.05),白芨组的细胞凋亡率(26.01±2.10)较模型组下降(p<0.05)。结论 白芨水煎剂能够抑制MC豚鼠结肠上皮细胞凋亡,从而改善MC。 |

| Objective To investigate the effects of bletilla decoction on ultrastructure of colonic mucosal epithelial cell in guinea pigs’ colon of MC caused by anthraquinone herbs. Methods 40 guinea pigs were randomly divided into control (n=8), MC model (n=16) and Bletilla (n=16) groups. The model group and the bletilla group were induced by intragastric administration of rhubarb at a dose of 6g/kg·d for 60 consecutive days. After 2 weeks, the animals in the bletilla group were treated with 10% bletilla decoction enema for three times a week while the model group were treated with NS enema. Melatonin staining, ultrastructural changes and flow cytometry were observed in colonic epithelial cells of guinea pigs. Results Compared with the normal group and the bletilla group, melatonin staining of guinea pigs in the model group showed positive changes, and the range was obviously larger than that in bletilla group. The model group could find different sizes of vacuolar change in most epithelial cells’ cytoplasm and apoptotic epithelial cells sametimes, while the bletilla group could find a little mitochondrion slight vacuolar change, and epithelial cell apoptosis is not obvious. Compared with the normal group(13.73±2.73%), the apoptosis rate of colonic epithelial cells in guinea pigs in the model group (35.27±1.21)was increased (p<0.05), and the apoptosis rate of Bletilla striata group(26.01±2.10) was lower than that of the model group (p<0.05). Conclusion Bletilla decoction can improve Melanosis Coli Caused by inhibiting the apoptosis of colon epithelial cells in MC guinea pigs. |
