| 目的 探讨桃叶珊瑚苷治疗骨质疏松的作用及可能的机制,为临床治疗骨质疏松提供实验基础。方法 取48只健康雌性Wistar大鼠,随机分为桃叶珊瑚苷高剂量组(10 mg/kg)、桃叶珊瑚苷低剂量组(5 mg/kg)、模型组(等量生理盐水)、假手术组(等量生理盐水)4组,每组12只。除假手术组外,其余3组大鼠麻醉后切除双侧卵巢构建骨质疏松模型。各组大鼠连续给药8周后处死,收集血清样本,采用酶联免疫测试方法(ELISA)检测血清中碱性磷酸酶(ALP)与骨钙素(BGP)含量;取大鼠左侧股骨测定骨密度;观察胫骨组织形态改变;取大鼠右侧股骨测荧光定量PCR检测骨组织中OPG、RANK、RANKL mRNA表达。结果 ELISA结果显示,与假手术组比较,模型组ALP、BGP含量差异有统计学意义(P<0.01) ,桃叶珊瑚苷高剂量组ALP、BGP含量降低与模型组比较差异有统计学意义(P<0.05)。与模型组比较,高剂量组大鼠骨密度值显著升高(P<0.01),低剂量组骨密度值升高,差异有统计学意义(P<0.05)。骨组织形态发现,桃叶珊瑚苷高剂量组骨小梁结构紧密, 连续性较好,优于模型组。桃叶珊瑚苷高剂量组能提高了骨组织中OPG mRNA表达,降低了RANK mRNA的表达。结论 桃叶珊瑚苷能够促进骨形成,减缓骨丢失,其作用机制可能通过 OPG/RANKL/RANK系统发挥作用。 |

| Objective To investigate the effect and possible mechanism of aucubin in the treatment of osteoporosis, and to provide an experimental basis for the clinical treatment of osteoporosis. Methods Forty-eight healthy female Wistar rats were randomly divided into aucubin high-dose group (10 mg/kg), aucubin low-dose group (5 mg/kg), model group (equal saline), There are 4 groups in the sham operation group (equal amount of normal saline), with 12 rats in each group. Except for the sham operation group, the rats in the other 3 groups were anesthetized and removed bilateral ovaries to construct an osteoporosis model. The rats in each group were sacrificed after 8 weeks of continuous administration. Serum samples were collected, and the serum alkaline phosphatase (ALP) and osteocalcin (BGP) contents were detected by enzyme-linked immunoassay (ELISA); the left femur of the rats was taken The bone density was measured; the morphological changes of the tibia were observed; the right femur of the rat was taken for fluorescence quantitative PCR to detect the expression of OPG, RANK, RANKL mRNA in the bone tissue. Results ELISA results showed that compared with the sham operation group, the difference in ALP and BGP content in the model group was statistically significant (P<0.01), and the reduction in ALP and BGP content in the high-dose aucubin group was statistically significant compared with the model group ( P<0.05). Compared with the model group, the bone mineral density of rats in the high-dose group was significantly increased (P<0.01), and the bone mineral density in the low-dose group was increased, and the difference was statistically significant (P<0.05). The morphology of bone tissue found that the trabecular bone structure of the aucubin high-dose group was tighter and the continuity was better, which was better than that of the model group. The aucubin high-dose group can increase the expression of OPG mRNA in bone tissue and decrease the expression of RANK mRNA. Conclusion Aucubin can promote bone formation and slow down bone loss, and its mechanism of action may play a role through the OPG/RANKL/RANK system. |
