| 目的：通过网络药理学探讨运脾化浊膏干预代谢综合征的作用机制。方法：筛选并重合运脾化浊膏与代谢综合征的靶点，于String11.5平台构建蛋白质互作网络，于Metascape平台进行GO，KEGG分析，通过Cytoscape3.8.2构建出“药物-四气五味归经”、“药物-成分-作用靶点”、“药物-疾病PPI”、“药物-成分-疾病-通路”等网络图并加以分析网络图与作用机制。结果：运脾化浊膏内有效成分99种，作用靶点261个；挖掘疾病数据库获得代谢综合征疾病靶点2464个，共同靶点261个，筛选出核心靶点26个。经Cytoscape3.8.2预测其主要活性成分可能为β-谷固醇及豆甾醇等，分析GO与KEGG通路，运脾化浊膏可通过结合DNA结合转录因子、磷酸酶、染色质、转录因子等，并调节信号受体激活剂、核受体、配体激活转录因子等活性来改善MS相关症状，及主要通路可能与AGE-RAGE信号通路、IL-17信号通路、Toll样受体信号通路相关。结论：初步探究运脾化浊膏治疗代谢综合征的作用机制，其作用通路可能为AGE-RAGE信号通路、IL-17信号通路、Toll样受体信号通路。 |

| Objective:To explore the mechanism of intervention of Yunpi Huazhuo Ointment on metabolic syndrome through network pharmacology.Methods:Screening and overlapping targets of pihuazhuan ointment and metabolic syndrome,protein interaction network was constructed on String11.5 platform,GO and KEGG analysis was performed on Metascape platform.Through Cytoscape3.8.2,the network diagrams of "drug-four qi and five flavors pathway","drug-component-action target","drug-disease PPI"and"drug-component-disease-pathway"are constructed and analyzed.Results:There are 99 kinds of active components and 261 targets in Yunpi Huazhuo ointment.A total of 2464 metabolic syndrome disease targets,261 common targets and 26 core targets were obtained by mining the disease database.According to Cytoscape3.8.2,it is predicted that the main active components may be β -sitosterol and stigmasterol,etc.By analyzing the GO and KEGG pathways,Yunpihuazhuo extract can improve MS related symptoms by binding DNA to transcription factors, phosphatase,chromatin and transcription factors,and regulating the activities of signal receptor activator,nuclear receptor and ligand-activated transcription factors.And the main pathways may be related to age-rage signaling pathway,IL-17 signaling pathway and Toll-like receptor signaling pathway. Conclusion:To preliminarily explore the mechanism of yunpi Huazhuo Ointment in the treatment of metabolic syndrome, its pathway may be age-rage signaling pathway,IL-17 signaling pathway and Toll-like receptor signaling pathway. |
