      目的 观察阿魏酸对去卵巢大鼠骨微结构改变的作用。方法 2月龄雌性大鼠40只,按随机数字表法分为假手术组、模型组和阿魏酸低、高剂量组,每组10只。除假手术组外,模型组和阿魏酸低、高剂量组均通过摘除大鼠双侧卵巢建立骨质疏松模型,分别采用等量生理盐水和20、80 mg/(kg·d)阿魏酸灌胃,干预12周后,处死各组大鼠获取大鼠血清检测骨代谢指标,用Micro-CT检查记录大鼠股骨骨密度和骨微结构,切取干骺端骨质行HE染色,观察骨组织切片。结果 病理切片显示,模型组大鼠骨小梁变薄稀疏,髓腔增大;阿魏酸组大鼠骨小梁数目(Tb.N)和骨连接增加,骨小梁间隙变小。与模型组比较,阿魏酸低、高剂量组大鼠骨密度增加[(0.176±0.014) g/cm2、(0.198±0.027) g/cm2比(0.153±0.026) g/cm2,P<0.05]、皮质骨骨小梁体积比率(BV/TV)增加[(34.521±6.364)%、(39.483±10.586)%比(26.357±8.243)%,P<0.05]、骨小梁厚度(Tb.Th)增加[(0.116±0.031) mm、(0.129±0.028) mm比(0.092±0.012) mm,P均<0.05],骨小梁数目(Tb.N)增多[(2.684±0.087) 1/mm、(2.851±0.102) 1/mm比(2.452±0.121) 1/mm,P<0.05],骨小梁间隙(Tb.Sp)缩小[(0.406±0.029) mm、(0.326±0.034) mm比(0.518±0.031) mm,P<0.05];与模型组比较,阿魏酸低、高剂量组大鼠血钙浓度[(2.32±0.24) mmol/L、(2.48±0.19) mmol/L比(2.42±0.32) mmol/L]、血磷浓度[(1.72±0.28) mmol/L、(1.64±0.24) mmol/L比(1.58±0.32) mmol/L]和碱性磷酸酶(ALP)浓度[(68.5±20.3) U/L、(70.2±24.6) U/L比(61.4±18.8) U/L],含量均相近(P>0.05),骨钙素(OC)浓度降低[(3.894±0.984) ng/mL、(3.252±1.028) ng/mL比(5.246±0.824) ng/mL,P<0.05]。结论 阿魏酸可减少去势骨质疏松大鼠骨量丢失,改善骨微结构。

      To observe the effect of ferulic acid on the changes of bone microstructure in ovariectomized rats. Methods Forty 2-month-old female rats were randomly divided into sham operation group, model group and low-dose and high-dose ferulic acid groups, with 10 rats in each group. Except for the sham-operated group, the model group and the low-dose and high-dose ferulic acid groups established osteoporosis models by removing the bilateral ovaries of the rats. After 12 weeks of intervention, rats in each group were sacrificed to obtain rat serum to detect bone metabolism indicators, and Micro-CT examination was used to record the bone mineral density and bone microstructure of rat femur, and metaphyseal bone was excised for HE staining. Bone tissue slices. Results Pathological sections showed that the trabecular bone became thinner and sparser and the medullary cavity increased in the model group; the number of trabecular bone (Tb.N) and bone connection in the ferulic acid group increased, and the trabecular bone gap became smaller. Compared with the model group, the bone mineral density of the rats in the low-dose and high-dose ferulic acid groups increased [(0.176±0.014) g/cm2, (0.198±0.027) g/cm2 to (0.153±0.026) g/cm2, P<0.05 ], cortical bone trabecular volume ratio (BV/TV) increased [(34.521±6.364)%, (39.483±10.586)% to (26.357±8.243)%, P<0.05], trabecular thickness (Tb.Th) increased [(0.116±0.031) mm, (0.129±0.028) mm to (0.092±0.012) mm, P<0.05], and the number of trabecular bone (Tb.N) increased [(2.684±0.087) 1/mm , (2.851±0.102) 1/mm to (2.452±0.121) 1/mm, P<0.05], trabecular bone space (Tb.Sp) decreased [(0.406±0.029) mm, (0.326±0.034) mm to (0.518±0.031) mm, P<0.05]; compared with the model group, serum calcium concentration of rats in low-dose and high-dose ferulic acid groups [(2.32±0.24) mmol/L, (2.48±0.19) mmol/L to (2.42±0.32) mmol/L], blood phosphorus concentration [(1.72±0.28) mmol/L, (1.64±0.24) mmol/L to (1.58±0.32) mmol/L] and alkaline phosphatase (ALP) concentration [(68.5 ±20.3) U/L, (70.2±24.6) U/L to (61.4±18.8) U/L], content were similar (P>0.05), and osteocalcin (OC) concentration decreased [(3.894±0.984) ng/mL, (3.252±1.028) ng/mL to (5.246±0.824) ng/mL,P<0.05]. Conclusion Ferulic acid can reduce bone loss and improve bone microstructure in castrated osteoporotic rats
