| 目的 探究葛根芩连汤对葡聚糖硫酸钠(DSS)诱导小鼠溃疡性结肠炎(UC)的抗炎作用。方法 34只SPF级C57BL/6小鼠按随机数字表法分为空白组8只、模型组10只、柳氮磺吡啶组(260mg/kg)8只和葛根芩连汤组(390mg/kg)8只。除空白组外,其余各组小鼠自由饮用2.5% DSS建立UC小鼠模型,建模成功后,柳氮磺吡啶组、葛根芩连汤组小鼠每天给予相应药物灌胃,其余组给予等体积(10ml/kg)生理盐水。每天观察小鼠体质量、大便性状、神态等一般情况的变化,并参照疾病活动指数(DAI)表进行评分。实验结束时采集结肠组织标本,采用苏木素-伊红(HE)染色观察各组小鼠结肠组织病理改变进行结肠组织学评分,酶联免疫吸附测定(ELISA)检测小鼠血清肿瘤坏死因子-α(TNF-α)和白细胞介素-1β(IL-1β)浓度。 结果 与空白组比较,模型组小鼠体质量明显下降,出现腹泻、血便,DAI评分[(1.43±0.47)分比(0.00±0.00)分,P<0.05]、组织学评分[(6.80±3.99)分比(0.00±0.00)分,P<0.05]、血清中IL-1β[(49.14±18.52)pg/mL比(28.7±5.68)pg/mL,P<0.05]和TNF-α[(405.77±82.06)pg/mL比(295.32±60.93)pg/mL,P<0.05]浓度显著上升,结肠明显缩短[(5.40±0.41)cm比(7.07±0.61)cm,P<0.05]。经葛根芩连汤干预后,葛根芩连汤组与模型组相比小鼠体质量、大便情况等明显改善,DAI评分[(1.24±0.92)分比(1.43±0.47)分,P>0.05]、组织学评分[(5.00±4.93)分比(6.80±3.99)分,P>0.05]、血清TNF-α[(362.36±71.37)pg/mL比(405.77±82.06)pg/mL,P<0.05]、IL-1β[(46.73±10.02)pg/mL比(49.14±18.52)pg/mL,P>0.05]浓度降低,结肠组织形态和长度改善[(5.60±0.54)cm比(5.40±0.41)cm,P>0.05],与柳氮磺吡啶组小鼠之间无统计学差异(P>0.05)。 结论 葛根芩连汤能一定程度改善DSS诱导的UC小鼠结肠组织病理情况,其机制可能与下调炎性因子TNF-α的表达有关。 |

| Objective To explore the anti-inflammatory effect of Gegen Qinlian decoction on DSS-induced ulcerative colitis(UC). Methods 34 C57BL/6 mice were randomly divided into four group: normal contrast (n=8, NC group), dextran sulfate sodium (n=10, DSS group), sulfasalazine (260mg/kg, n=8, SASP group), Gegen Qinlian decoction(390mg/kg, n=8, GD group). Except NC group, mice in other group were fed with 2.5% DSS solution establish acute UC model. Mice in SASP group or GD group were given sulfasalazine or Gegen Qinlian decoction via oral gavage. The changes of body weight, fecal traits, spirt and other general conditions of the mice were observed every day, and scored by referring to the disease activity index(DAI) table. At the end of the experiment, colon tissue samples were collected, and the pathological changes of colon tissues were observed by Hematoxylin-Eosin (HE) staining for colon histological injury score(HIS). Serum levels of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) were determined by enzyme-linked immunosorbent assay (ELISA). Result Compared with NC group, the body weight of mice in DSS group significantly were decreased, and DAI score[(1.43±0.47) vs. (0.00±0.00); P<0.05], HIS score[(6.80±3.99) vs. (0.00±0.00); P<0.05] and the serum level of inflammatory factor IL-1β[(49.14±18.52)pg/mL vs. (28.7±5.68)pg/mL; P<0.05] and TNF-α[(405.77±82.06)pg/mL vs. (295.32±60.93)pg/mL; P<0.05] were significantly increased, as well as colon length was significantly shortened[(5.40±0.41)cm vs. (7.07±0.61)cm; P<0.05]. After GD intervention, compared with DSS group, the body weight and stool condition of mice in GD group were improved, and DAI score[(1.24±0.92) vs. (1.43±0.47); P>0.05], HIS score[(5.00±4.93) vs. (6.80±3.99); P>0.05], serum TNF-α [(362.36±71.37)pg/mL vs. (405.77±82.06)pg/mL; P<0.05]and IL-1β[(46.73±10.02)pg/mL vs. (49.14±18.52)pg/mL; P>0.05] concentration were decreased, the morphology and length of colon[(5.60±0.54)cm vs. (5.40±0.41)cm, P>0.05] tissue were improved. There was no significant difference between SASP group and GD group (P>0.05). Conclusion GD could improve DSS-induced UC mice to a certain extent by down-regulate the serum level of inflammatory factor TNF-α. |
