      目的:检测酪氨酸激酶受体RON和MET蛋白在三阴性乳腺癌(TNBC)组织中的表达,并分析其与临床病理分期、组织分级、淋巴结转移以及患者生存期的关系,以探究这两个生物标志在TNBC临床病理诊断及病人预后判断中的应用价值。方法:收集130例三阴性乳腺癌患者手术切除的癌组织,应用免疫组织化学染色法检测癌组织RON和MET蛋白的表达水平,并进行量化积分。结果:在三阴性乳腺癌组织中存在广泛的RON和MET异常表达,其中RON的阳性表达率为73.08%,MET的阳性表达率高达77.69%,且RON和MET过表达的患者可分别达到38.46%和44.62%;二者的表达水平具有相关性(P<0.05)。MET蛋白表达与乳腺癌组织学分级相关(P<0.05)。通过生存期分析,证实癌组织RON或MET过表达的患者生存期明显缩短。结论:癌组织中异常表达的RON和MET参与三阴性乳腺癌患者的病理进展,其过表达可作为评估预后不良的重要生物标志物。

      Objective To detect the expression of tyrosine kinase receptor RON and MET protein in triple-negative breast cancer (TNBC) tissues, and to analyze their relationship with related clinical indicators, in order to explore the value of these two indicators of clinical pathological diagnosis and the patient prognosis judgment. Methods The surgically resected cancer tissues of 130 triple-negative breast cancer patients were collected. Immunohistochemically staining was used to detect the expression levels of RON and MET proteins in cancer tissues. The Envision method was used to quantitatively integrate the expression levels of RON and MET. Results There are extensive abnormal expressions of RON and MET in triple-negative breast cancer tissues. The positive expression rate of RON is 73.08%, the rate of MET is as high as 77.69%, and the overexpression of RON and MET can reach 13.62% and 12.31% respectively; The expression of them is correlated (P<0.05) and the expression of MET protein is correlated with the histological grade of breast cancer (P<0.05). Through survival analysis, it was confirmed that the survival time of patients with co-overexpression of RON and MET in cancer tissues was significantly shortened. Conclusion The abnormally expressed RON and MET in cancer tissues are involved in the pathological progression of triple-negative breast cancer patients, and their co-overexpression can be used as an important biomarker for poor prognosis.
