| 目的 探讨五味子醇甲(Schisandrin A,SCH A)通过Aβ25-35诱导PC12细胞损伤对氧化应激及炎症因子的影响。方法 CCK8、流式细胞仪联合检测,设立20 μmol/L Aβ25-35诱导PC12建立细胞损伤模型。MTT、流式细胞仪确立10μg/mL SCH A为本次实验药物剂量。实验分为空白组、模型组、SCH A组。试剂盒检测超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)表达水平;Western Blot检测白介素-1β(IL-1β)、白介素-6(IL-6)、肿瘤坏死因子(TNF-α)蛋白水平。结果 与模型组比较,SCH A组能提高SOD、GSH水平,降低MDA水平,有显著差异(P<0.01);能降低IL-1β、IL-6、TNF-α蛋白表达水平,有显著差异(P<0.01)。结论SCH A可能通过降低Aβ25-35诱导PC12细胞的炎性反应,减轻氧化应激,从而发挥细胞保护作用。 |

| Objective To investigate the effects of (Schisandrin A,SCH A)Son oxidative stress and inflammatory factors of PC12 cells induced by Aβ25-35. Methods CCK8 was combined with flow cytometry to induce PC12 with 20 μmol/L Aβ25-35 to establish cell damage model. MTT and flow cytometry determined 10μg/mL SCH A as the dose of this experiment. The experiment was divided into blank group, model group and SCH A group. The expression levels of superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione (GSH) were detected by the kit. Western Blot analysis of interleukin-1 β (IL-1β), interleukin-6 (IL-6), tumor necrosis factor (TNF-α) protein levels. Results SCH A group could increase the level of SOD and GSH and decrease the level of MDA, with significant difference (P<0.01). SCH A could decrease the expression levels of IL-1β, IL-6 and TNF-α with significant difference (P<0.01). Conclusion SCH A may play A protective role in PC12 cells by reducing the inflammatory response induced by Aβ25-35 and alleviating oxidative stress. |
