| 目的 运用网络药理学探究大黄治疗卒中后抑郁(Post-Stroke Depression,PSD)机制。方法 检索TCMSP中药系统药理学数据库与分析平台,筛选出大黄潜在活性成分及靶点,并整理对应药活性成分及潜在靶点信息,然后把获得的靶点蛋白名通过UniProt数据库转换为基因名。通过GeneCards数据库、OMIM数据库、DRUGBANK数据库筛选卒中后抑郁疾病相关靶点。通过GO和 KEGG进行相关分析并完成数据可视化。结果 得到大黄潜在活性成分16个,其中可获得数据成分13个,疾病靶点5167个,药物-疾病交集潜在作用靶点72个,靶点生物功能主要涉及对脂多糖介质反应及介导其通路、激活MAPK活动、对缺氧反应、蛋白质水解、基因表达等,且通过调节TNF、MAPK、Toll样受体、NOD样受体、血清素、多巴胺等信号通路达到延缓卒中后抑郁发生发展的作用。结论 大黄可能通过TNF、MAPK、Toll样受体、NOD样受体、血清素、多巴胺等通路,调节机体内多种蛋白酶及神经递质活性以延缓卒中后抑郁进展。 |

| Objective:Use network pharmacology to explore the mechanism of rhubarb in the treatment of post-stroke depression. Methods:Search the TCMSP traditional Chinese medicine system pharmacology database and analysis platform, screen out the potential active ingredients and targets of rhubarb, and sort out the corresponding active ingredients and potential target information, and then convert the obtained target protein names into gene names through the UniProt database. Screening related targets for post-stroke depression diseases through GeneCards database, OMIM database, and DRUGBANK database. Perform relevant analysis and complete data visualization through GO and KEGG. Results:obtained 16 potential active components of rhubarb, of which 13 data components can be obtained, 5167 disease targets, 72 drug-disease intersection potential targets. The biological functions of the targets mainly involve the response to lipopolysaccharide mediators and mediate its pathways, Activate MAPK activity, response to hypoxia, proteolysis, gene expression, etc., and delay the development of post-stroke depression by regulating signal pathways such as TNF, MAPK, Toll-like receptors, FoxO, NOD-like receptors, serotonin, and dopamine effect. Conclusions:Rhubarb may regulate the activities of various proteases and neurotransmitters in the body through TNF, MAPK, Toll-like receptors, FoxO, NOD-like receptors, serotonin, dopamine and other pathways to delay the progression of post-stroke depression. |
