      目的 研究血根碱(sanguinarine,San)在大肠癌细胞增殖和迁移中的作用及其作用机理。 方法 目的探讨不同剂量 San (2,4,8μ?mol/L)对大肠癌细胞系(HCT8、SW620)进行治疗,并应用细胞计数仪CCK-8及细胞克隆试验观察各组细胞的生长情况;用蛋白印迹的方法测定了相关蛋白的表达;建立HCT8皮下瘤体模型,采用10?mg/kg的 San液进行腹腔内灌注,以探讨其对结肠癌的抑制作用。 结果 CCK-8和细胞系的形成试验表明,San对大肠癌的增殖和增殖有一定的影响;细胞划痕试验表明,San会显著抑制大肠癌细胞的转移;免疫印迹试验表明,San能显著降低大肠癌组织中N-cadherin、PI3K、p-AKT和?mTOR的表达,使E-cadherin蛋白的表达增加;体内试验表明,San液对皮下肿瘤的生长有显著的抑制作用。 结论 血根碱能抑制大肠癌细胞的增殖和迁移,具有一定的体内外抑瘤活性,并且能调节结肠癌细胞的PI3K/AKT信号通路。
英文摘要:
      Objective: To explore the inhibited function and possibly mechanism of sanguinarine (San) to colon cancer. Methods: Colon cancer cell lines (HCT8 and SW620) were treated with different concentrations of San (2, 4, 8μmol/L). Cell proliferation was detected by cell counting kit (CCK-8) and cell clone formation, and cell migration was detected by wound-healing assay. The expressions of related proteins were detected by Western blot. HCT8 subcutaneous tumor mouse model was constructed and San solution was injected intraperitoneally at a dose of 10mg/kg to observe the inhibitory ability of San on subcutaneous tumor. Results: Cck-8 and clone formation assay showed that San had a certain effect on the proliferation and proliferation of colorectal cancer. The cell wound-healing assay indicated that hat San could significantly inhibit the metastasis of colorectal cancer cells. Western blot showed that San significantly decreased the expression of N-cadherin, PI3K, p-Akt and mTOR, and increased the expression of E-cadherin protein in colorectal cancer. In vivo, In vivo, it has been found that San solution had a significant inhibitory effect on the growth of subcutaneous tumors. Conclusion: Sanguinarine can inhibit the proliferation and migration of colorectal cancer cells, and has certain antitumor activity in vitro and in vivo, thus showing anti-tumor effects, and this effect is related to its regulation of PI3K/AKT.
