| :[目的]通过网络药理学方法,分析浙贝-甘草药对干预非小细胞肺癌(NSCLC)的可能信号通路;结合体内实验,研究浙贝-甘草药对调控细胞核因子-κB(NF-κB)信号通路干扰NSCLC的相关机制。[方法]1.通过网络药理学研究,获得浙贝-甘草药对的活性化学成分及潜在作用靶点、收集NSCLC的相关靶标基因。运用富集分析等方法,分析浙贝-甘草药对干预NSCLC的可能信号通路及作用靶点。2.通过体内相关实验评价浙贝-甘草药对对移植瘤生长的作用。 [结果]1.浙贝-甘草药对中含有槲皮素、β-谷甾醇、山奈酚、甘草醇、谷甾醇为主要有效成分,等具有抗肿瘤、抗炎、调节免疫作用的有效活性成分,其干预NSCLC可能与调控β转化生长因子(TGF-β)信号通路、p53信号通路、NF-κB信号通路等相关。2.体内实验显示,与模型组相比,药对等效剂量组瘤体积减少[(358.74±50.54) mm3比(743.62±125.95) mm3, P<0.01],瘤重减轻[(0.28±0.05)g比(0.66±0.16)g, P<0.01],HE半定量评分升高[(3.20±0.45)比(0.20±0.45), P<0.01],Toll样受体4(TLR-4)、细胞核因子-κB 1(NF-κB1)、细胞核因子-κB p65(RelA) 蛋白表达降低[TLR-4:(0.662±0.054)比(1.000±0.085); NF-κB1:(0.457±0.042)比(1.000±0.084); RelA:(0.580±0.053)比(1.000±0.089),P均<0.01]。 [结论]浙贝-甘草药对可明显抑制瘤体细胞增殖,诱导细胞凋亡,其机制可能与调控NF-κB信号通路有关。 |

| Objective To analyze the possible signaling pathways of Zhebei-Gancao herb pair in the intervention of Non-Small Cell Lung Cancer ( NSCLC ) by network pharmacology. Combined with in vivo experiments, the mechanism of Zhebei-Gancao drug pair regulating NF-κB signaling pathway to interfere with NSCLC was studied. Methods 1.Through the study of network pharmacology, the active chemical components and potential targets of Zhebei-Gancao herb pair were obtained, and the related target genes of NSCLC were collected. Enrichment analysis and other methods were used to analyze the possible signaling pathways and targets of Zhebei-Gancao drug pair in the intervention of NSCLC. The effect of Zhebei-Gancao herb pair on the growth of transplanted tumor was evaluated by in vivo experiments. Result 1.Zhebei-Gancao drug pair contains quercetin, β-sitosterol, kaempferol, liquiritin, sitosterol as the main active ingredients, and other effective active ingredients with anti-tumor, anti-inflammatory, and immunoregulatory effects. Its intervention in NSCLC may be related to the regulation of TGF-β signaling pathway, p53 signaling pathway, NF-κB signaling pathway and so on. 2. In vivo experimental results showed that compared with the model group, the volume of tumors in the equivalent dose group was reduced [(358.74±50.54) mm3 vs. (743.62±125.95) mm3, P<0.01], tumor weight was reduced [(0.28±0.05)g vs. (0.66±0.16) g, P<0.01], and the HE semiquantitative score was increased [ 3.20±0.45 vs. 0.20±0.45, P <0.01], Toll-like receptor-4(TLR-4), Nuclear factor-κB 1(NF-κB1), Nuclear factor-κB p65(RelA) protein expression decreased [TLR-4: (0.662±0.054) vs. (1.000±0.085); NF-κB1: (0.457±0.042) vs. (1.000±0.084); RelA: (0.580±0.053) vs. (1.000±0.089), P <0.01].Conclusion Zhebei-Gancao herb pair can significantly inhibit the proliferation of tumor cells and induce apoptosis, and its mechanism may be related to the regulation of NF-κB signaling pathway. |
