| 目的 观察胡柚皮黄酮对棕榈酸诱导的肝细胞脂肪变性的影响。方法 棕榈酸诱导HepG2细胞脂肪变性,不同浓度胡柚皮黄酮干预后,采用油红O染色法观察肝细胞脂肪变性;荧光探针法观测细胞内活性氧水平及线粒体膜电位变化;免疫印迹法检测Beclin-1、LC3及P62蛋白表达。结果 棕榈酸诱导后,HepG2细胞内脂滴和活性氧含量明显增高;胡柚皮黄酮处理后,细胞内脂滴和活性氧均明显降低,线粒体膜电位恢复,LC3和Beclin-1蛋白表达上调,P62蛋白表达下调。结论 胡柚皮黄酮能改善肝细胞脂肪变性和氧化应激反应,机制可能与其调控线粒体自噬,维持线粒体功能稳定有关。 |

| BSTRACT Objective To investigate the effect of Pure total flavonoids from Citrus(PTFC) on palmitic acid induced hepatocyte steatosis. Methods HepG2 cells were induced by palmitic acid to established hepatic steatosis cell model. The protective effects of PTFC on hepatic steatosis were observed by oil red O staining. Fluorescence probe method was used to detect intracellular reactive oxygen species(ROS) levels and mitochondrial membrane potential; The expression of Beclin-1, LC3 and P62 proteins were detected by western blot. Results After induction with palmitic acid, the content of lipid droplets and ROS in HepG2 cells increased Compared with the control group. the lipid droplets and ROS in the cells were significantly decreased, and the mitochondrial membrane potential was restored. The expression of LC3 and Beclin-1 proteins were increased, and the expression of P62 protein was downregulated after the treatment with PTFC. Conclusion PTFC improves hepatic steatosis and oxidative stress response by regulating mitochondrial autophagy and maintaining mitochondrial functional stability. |
