      目的 观察苦藏花素协同吉非替尼对人肺癌A549细胞增殖的抑制作用。方法 采用Cell Counting Kit-8(CCK8)检测单独用药(苦藏花素、吉非替尼)和联合用药(苦藏花素+吉非替尼)组不同浓度药物处理24,48,72h对A549细胞增殖的抑制作用;使用两药相互作用系数(Coefficient of Drug in Interaction, CDI)来评价吉非替尼和苦藏花素的相互作用性质。结果 苦藏花素和吉非替尼对A549细胞的增殖均有不同程度的抑制作用,在一定范围内与药物浓度呈正相关,且存在时间依赖性;其中,160ug/ml剂量的苦藏花素和低剂量吉非替尼(10ug/ml)协同抑制作用,可以达到高剂量吉非替尼(20ug/ml)单独用药的抑制作用。160ug/ml的苦藏花素和一定浓度范围内的吉非替尼CDI值小于1。结论 苦藏花素联用半量吉非替尼可以达到全量使用吉非替尼的抑制效果,提示苦藏花素联用吉非替尼可以协同抑制肺癌细胞A549的增殖。
英文摘要:
      Objective To observe the inhibitory effect of picrocrocin combined with gefitinib on the proliferation of human lung cancer A549 cells.Methods CCK8 method was used to detect the inhibition of proliferation of A549 cells at different concentrations in the groups of single drug (picrocrocin and gefitinib) and combined drug (picrocrocin + gefitinib) at 24, 48 and 72 hours. Coefficent of Drug in Interaction (CDI) was used to evaluate the Interaction Properties between picrocrocin and gefitinib.Results Both picrocrocin and gefitinib inhibited the proliferation of A549 cells in varying degrees, which was positively correlated with the drug concentration in a certain range and in a time-dependent manner. Among them, the synergistic effect of 160ug/ml dose of picrocrocin and low-dose gefitinib (10ug/ml) can achieve the inhibitory effect of high-dose gefitinib (20ug/ml) alone.The CDI value of picrocrocin in160ug/ml and gefitinib within a certain concentration range is less than 1.Conclusion Picrocrocin combined with half dose gefitinib can achieve the same inhibitory effect as full dose gefitinib, suggesting that picrocrocin combined with gefitinib can synergistically inhibit the proliferation of lung cancer cell A549.
