| 目的 探讨红景天对肺血管通透性和BMP9表达的影响。方法 将40只小鼠平均分为假手术组、模型组和低、中、高剂量红景天组,采用盲肠结扎-穿孔法造模,假手术组开腹后关腹,红景天组分别用40mg/kg、80mg/kg、160mg/kg灌胃5天,处死后用小鼠肺组织行湿干重比、血管渗漏检测和免疫组化、Western blotting法检测BMP9表达水平。使用LPS诱导HUVEC细胞形成内皮细胞炎性损伤模型,用CCK-8法检测细胞活性,筛选出红景天浓度(25、50、100μg/ mL)。将细胞分为对照组(无LPS诱导)、模型组、高中低浓度红景天组(25、50、100μg/ mL),应用qPCR、Western blotting、免疫荧光技术检测BMP9表达情况。结果 各组小鼠湿干重比、血管渗漏检测结果均有统计学差异(P<0.01)。Western blotting检测各组小鼠肺组织BMP9表达(F=181,P<0.001)和免疫组化检测五组小鼠肺组织BMP9表达水平比较,差异均具有统计学意义。qPCR法检测五组HUVEC细胞BMP9 RNA扩增值差异(F=36.8,P<0.001),Western blotting检测五组细胞BMP9蛋白表达差异(F=107,P<0.001)和免疫荧光法检测各组细胞间BMP9免疫荧光强度分别为(41.878、10.062、21.875、24.768、32.008),差异均具有统计学差异。结论 红景天具有肺血管保护作用,其机制可能与增加BMP9表达有关。 |

| Objective To investigate the effects of Rhodiola rosea on pulmonary vascular permeability and BMP9 expression. Methods 40 mice were divided into sham operation group, model group and low, medium and high dose Rhodiola rosea groups. Cecal ligation and perforation were used to make models. Sham operation group was closed after laparotomy, and Rhodiola rosea group was given 40mg/kg, 80mg/kg and 160mg/kg using gavage administration for 5 days respectively. The expression level of BMP9 in the lung tissues of mice was detected by immunohistochemistry and Western blotting the same as wet-dry weight ratio and vascular leakage were detected. HUVEC cells were induced by LPS to form an endothelial cell inflammatory injury model. The cell activity was detected by CCK-8 method, and the concentration of rhodiola rosea (25, 50, 100μg/ mL) was screened. The cells were divided into control group (without LPS induction), model group, and high, middle and low concentration rhodiola rosea group (25, 50, 100μg/ mL). The expression of BMP9 was detected by qPCR, Western blotting, and immunofluorescence techniques. Results There were statistical differences in wet-dry weight ratio and blood vessel leakage among all groups (P<0.01). The expression of BMP9 in lung tissues of mice in each group was detected by Western blotting (F=181, P<0.001) and in lung tissues of mice in the five groups by immunohistochemistry, the differences were statistically significant. qPCR was used to detect the difference of BMP9 RNA proliferation in the five groups of HUVEC cells (F=36.8, P<0.001), and Western blotting was used to detect the difference of BMP9 protein expression in the five groups of cells (F=107, P<0.001).And the immunofluorescence intensity of BMP9 among all groups was 41.878, 10.062, 21.875, 24.768, 32.008,respectively.All results showed significant differences. Conclusions Rhodiola rosea has pulmonary vascular protective effect, and its mechanism may be related to the increase of BMP9 expression. |
