      目的 基于高迁移率族蛋白B1(HMGB1)/Toll样受体4(TLR4)/核因子-κB(NF-κB)信号通路研究枇杷叶提取物对急性肺损伤大鼠炎症因子的影响及潜在机制。方法 60只大鼠分为空白组(NC组)、模型组(ALI组)、阳性对照组(Dex组)、枇杷叶提取物低剂量组(EJLE-L组)、枇杷叶提取物中剂量组(EJLE-M组)、枇杷叶提取物高剂量组(EJLE-H组)各10只,ALI组、Dex组、EJLE-L组、EJLE-M组、EJLE-H组气管内均滴注2mg/kg脂多糖,Dex组腹腔注射5mg/kg地塞米松,EJLE-L组、EJLE-M组、EJLE-H组腹腔分别注射50、100、150mg/kg枇杷叶提取物,NC组注射5mg/kg生理盐水。观察大鼠肺组织病理组织学及肺组织细胞超微结构变化,比较各组大鼠肺组织湿重/干重值、外周循环、肺组织炎症因子白介素(IL)-1β、IL-6、肿瘤坏死因子(TNF)-α水平、HMGB1、TLR4、NF-κB累积光密度值(IOD)及蛋白表达量。结果 与NC组比较,ALI组湿重/干重、肺组织病理评分、外周循环/肺组织炎症因子水平、HMGB1、TLR4、NF-κB IOD值、表达量升高(P<0.05);与ALI组比较,Dex组、EJLE-L组、EJLE-M组、EJLE-H组湿重/干重、肺组织病理评分、外周循环/肺组织炎症因子水平、HMGB1、TLR4、NF-κB IOD值、表达量降低(P<0.05);与Dex组比较,EJLE-L组、EJLE-M组、EJLE-H组湿重/干重、肺组织病理评分、外周循环/肺组织炎症因子水平、HMGB1、TLR4、NF-κB IOD值、表达量降低(P<0.05);与EJLE-L组、EJLE-M组比较,EJLE-H组湿重/干重、肺组织病理评分、外周循环/肺组织炎症因子水平、HMGB1、TLR4、NF-κB IOD值、表达量降低(P<0.05)。结论 枇杷叶提取物可明显抑制急性肺损伤大鼠外周及肺部炎症,其可能经HMGB1/TLR4/NF-κB信号通路逆转肺损伤。

      Objective Based on high mobility group protein B1 (HMGB1)/Toll like receptor 4 (TLR4)/nuclear factor- κ B (NF- κB) Signal pathway research on the effects and potential mechanisms of loquat leaf extract on inflammatory factors in rats with acute lung injury. Methods Sixty rats were divided into blank group (NC group), model group (ALI group), positive control group (Dex group), loquat leaf extract low-dose group (EJLE-L group), Loquat leaf extract medium-dose group (EJLE-M group) and Loquat leaf extract high-dose group (EJLE-H group) with 10 rats each. ALI group, Dex group, EJLE-L group, EJLE-M group and EJLE-H group were infused with 2mg/kg LPS, Dex group was intraperitoneally injected with 5mg/kg dexamethasone, EJLE-L group, EJLE-M group and EJLE-H group were intraperitoneally injected with 50, 100 and 150mg/kg loquat leaf extract, respectively. The NC group was injected with 5mg/kg normal saline. The changes of lung histopathology and ultrastructure of lung cells were observed. The levels of wet weight (W)/dry weight (D), peripheral circulation, interleukin-1β, IL-6, tumor necrosis factor (TNF) -α, HMGB1, TLR4, NF-κB accumulated optical density (IOD) and protein expression in lung tissue of all groups were compared. Results Compared with NC group, wet weight/dry weight, pathological score of lung tissue, inflammatory factor level of peripheral circulation/lung tissue, HMGB1, TLR4, NF-κB IOD value and expression level in ALI group were increased (P < 0.05). Compared with ALI group, wet weight/dry weight, lung histopathologic score, peripheral circulation/lung inflammatory factor level, HMGB1, TLR4, NF-κB IOD value and expression of EJLE-H group were decreased in Dex group, EJLE-L group, EJLE-M group and EJLE-H group (P < 0.05). Compared with Dex group, wet weight/dry weight, lung tissue pathological score, peripheral circulation/lung inflammatory factor level, HMGB1, TLR4, NF-κB IOD value and expression level in EJLE-L, EJLE-M and EJLE-H groups were decreased (P < 0.05). Compared with EJLE-L group and EJLE-M group, wet weight/dry weight, lung tissue pathological score, peripheral circulation/lung inflammatory factor level, HMGB1, TLR4, NF-κB IOD value and expression level in EJLE-H group were decreased (P < 0.05). Conclusion Loquat leaf extract can significantly inhibit peripheral and pulmonary inflammation in rats with acute lung injury, and it may reverse lung injury through HMGB1/TLR4/NF-κB signaling pathway.
